Cortisol and α-amylase awakening response in children and adolescents with functional neurological (conversion) disorder

Jason Chung et al.

Summary: The study examined the salivary cortisol and alpha-amylase awakening responses in children with functional neurological disorder, finding a dysregulation of the hypothalamic-pituitary-adrenal axis. Patients with the disorder showed a decrease in cortisol awakening response, which was negatively correlated with adverse childhood experiences and subjective distress. Conversely, controls showed an increase in cortisol awakening response and no significant correlation with adverse childhood experiences.

Identification of biopsychological trait markers in functional neurological disorders

Samantha Weber et al.

Summary: Weber et al. provide evidence for a stress-diathesis model of functional neurological disorders, highlighting trauma history and reduced brain volume as risk factors. The study examines cortisol regulation and brain alterations in patients with functional neurological symptoms, finding a flatter cortisol awakening response and reduced amygdala and hippocampus volumes. These findings support the role of psychological stress as a risk factor and reduced brain volume as a biological trait marker for the disorder.

Potential role of FKBP5 single-nucleotide polymorphisms in functional seizures

Ali A. A. Asadi-Pooya et al.

Summary: The associations between FKBP5 single-nucleotide polymorphisms (SNPs) and functional seizures (FS) were investigated in this study. It was found that patients with FS and major depressive disorder (MDD) had different genotypes in rs9470080 and rs1360780 SNPs of FKBP5 compared to healthy controls. However, it seems that FKBP5 polymorphisms were not associated with FS in the absence of depression.

Linking gene expression patterns and brain morphometry to trauma and symptom severity in patients with functional seizures

Johannes Jungilligens et al.

Summary: Within stress-diathesis models, adverse life experiences increase the susceptibility to functional neurological symptoms through neuroplasticity effects. This study found that symptom severity was positively correlated with sexual trauma, emotional neglect, and threat to life experiences. Furthermore, lower gray matter volumes were associated with higher symptom severity, sexual trauma burden, and emotional neglect. The genes related to stress-related signaling and neurodevelopment were spatially correlated with these volumetric findings.

Where the genome meets the connectome: Understanding how genes shape human brain connectivity

Aurina Arnatkeviciute et al.

Summary: The integration of modern neuroimaging methods with genetically informative designs and data sheds light on the molecular mechanisms underlying the structural and functional organization of the human connectome. Studies investigating the genetic basis of human brain network structure and function through quantifying phenotypic heritability, identifying specific DNA variants, and analyzing correlations between imaging phenotypes and gene expression profiles show that anatomical connectivity is under stronger genetic influence than functional connectivity, with certain regions and connections being more genetically controlled than others. This combination of imaging and genetics can help understand how genes drive brain dysfunction in different clinical disorders.
