Increased hydrogel stiffness enhances migration and invasion capacity of breast cancer cells, as well as the expression of osteolytic factors. Animal models of cancer metastasis to the bone

Summary: Bone metastasis is a major cause of mortality in various tumors. Developing disease models is crucial for understanding the process of bone metastasis and devising effective therapies. Different translational models, including animal models, cell line injection models, bone implant models, and patient-derived xenograft models, have been developed. However, a comprehensive compendium on different bone metastatic cancers is currently lacking. This study compiles animal models derived from current experiments, mainly involving breast and prostate cancer, to enhance preclinical model development and promote bone metastasis treatment.

Induction of Reactive Bone Stromal Fibroblasts in 3D Models of Prostate Cancer Bone Metastases

Summary: This study aimed to establish a biologically relevant 3D in vitro model that mimics the cellular and molecular profiles of metastasis-associated fibroblasts (MAFs) found in vivo. Using 3D in vitro cell culture models, the bone-derived fibroblast cell line, HS-5, was treated with conditioned media from metastatic-derived prostate cancer cell lines, PC3 and MDA-PCa 2b, or mouse-derived fibroblasts 3T3. The resulting HS5-PC3 and HS5-MDA cell lines displayed alterations in expression levels and genomic profiles consistent with subpopulations of MAFs reported in vivo. The use of these engineered 3D models could provide insights into the novel biology regulating metastatic growth and the role of fibroblasts in the colonization process.

Development and characterisation of suitably bioengineered microfibrillar matrix-based 3D prostate cancer model for in vitro drug testing

Summary: In this study, newly developed microfibrillar matrices were fabricated as 3D scaffolds for prostate cancer model. These scaffolds showed improved cell retention and facilitated growth and deposition of cancer-specific extracellular matrix. The newly developed 3D model demonstrated increased resistance to the chemodrug compared to 2D culture.

Mechanoresponsive regulation of fibroblast-to-myofibroblast transition in three-dimensional tissue analogues: mechanical strain amplitude dependency of fibrosis

Summary: This study investigated the effect of mechanical loading on fibroblast phenotype and identified the ideal mechanical strain amplitude for promoting tissue regeneration without initiating myofibroblast differentiation. The results showed that moderate mechanical strain had anabolic effects on tissue regeneration, while higher strain amplitudes stimulated fibrotic tissue formation and myofibroblast differentiation.

Matrix stiffness regulates bone repair by modulating 12-lipoxygenase-me-diated early inflammation

Summary: The study demonstrated that low matrix stiffness can polarize macrophages into an anti-inflammatory phenotype, promoting the expression of anti-inflammatory cytokines and specialized pro-resolving lipid mediators beneficial for MSC osteogenesis. These findings indicate that matrix stiffness regulates bone repair by modulating 12-LOX-mediated early inflammation, suggesting a potential clinical application of SPMs in bone regeneration.
