Harmonization and standardization of nucleus pulposus cell extraction and culture methods

Summary: This study aims to standardize the extraction and expansion techniques for nucleus pulposus (NP) cells, in order to reduce variability between laboratories and improve comparability of results. The most commonly used methods for NP cell extraction, expansion, and re-differentiation were identified through a global survey. Recommended protocols for NP cell culture in different species were provided to support harmonization and cross-lab comparisons.

Relationship between altered knee kinematics and subchondral bone remodeling in a clinically translational model of ACL injury

Summary: This study used a rodent model of ACL injury to investigate the direct effect of ACL injury on joint kinematics and the development of PTOA. The results showed that knee flexion angle and subchondral bone microarchitecture were severely impacted after ACL injury and were associated with PTOA development.

The immune-privilege theory of cartilage tissue is not fully supported by clinical studies, and further research on cartilage immunology is needed for clinical advances.

Human primary muscle stem cells regenerate injured urethral sphincter in athymic rats

Summary: This study demonstrated the efficacy of human muscle stem cells in restoring internal urinary sphincter function in a preclinical animal model. The injection of muscle stem cells significantly improved peak bladder pressure and pressure rise in the treated animals compared to the control group. Minimal scarring was observed in the treated rats.

Summary: The study compared the effects of implanting allogeneic acellular skeletal muscle grafts in immunodeficient rats and immunocompetent rats, showing an increase in newly regenerated muscle fibers and higher macrophage staining in the immunodeficient rats. However, there were no significant differences in animal gait and muscle force recovery between the two groups. Ultimately, more research is needed to understand how host responses to biomaterials vary based on the animal model used.
