Summary: Exosomes secreted by healthy periodontal ligament stem cells (PDLSCs) promote osteogenic differentiation of PDLSCs derived from periodontitis tissue. Treatment with exosomes can accelerate bone formation in rat models of periodontitis. Mechanistically, exosomes suppress over-activation of the canonical Wnt signaling pathway to recover the osteogenic differentiation capacity of inflammatory PDLSCs. These findings suggest that MSC-derived exosomes are an effective and practical cell-free therapeutic for the treatment of periodontitis.

Effects of advanced platelet-rich fibrin combined with xenogenic bone on human periodontal ligament stem cells

Summary: This study investigated the effects of a mixture of advanced platelet-rich fibrin (A-PRF) and xenogenic bone substitute material (XBSM) on the proliferation and migration of human periodontal ligament stem cells (hPDLSCs). The results showed that the A-PRF-XBSM mixture continuously released growth factors and enhanced cell proliferation and migration. Therefore, the combination of A-PRF and XBSM may have potential advantages for periodontal tissue regeneration.

Summary: This study successfully improved the performance of L-PRF scaffold by using genipin as a crosslinker. The results showed that genipin-modified L-PRF had better biomechanical performance, slower biodegradation, good bioavailability, and sustained release of growth factors. In vitro and in vivo experiments demonstrated that 0.01% w/v and 0.1% w/v genipin-crosslinked L-PRF had good porous structure, significantly promoted cell proliferation, and enhanced bone regeneration.

Higher times of storage after centrifugation did not impact on cell content and the release of growth factors.

The results showed that LPPRF promoted more extensive cell migration and retained growth factors, while Mg ions released from the Mg ring significantly enhanced calcium deposition of preosteoblasts, indicating great potential for critical-size bone defect repair.
