Progesterone as an Anti-Inflammatory Drug and Immunomodulator: New Aspects in Hormonal Regulation of the Inflammation

Summary: The specific regulation of inflammatory processes by steroid hormones, particularly progesterone and progestins, has been actively studied in recent years. The anti-inflammatory effects of progesterone involve the inhibition of NF-kappa B and COX, as well as the regulation of T-cell activation, cytokine production, and immune tolerance. Progesterone and its derivatives can also inhibit proliferative signaling pathways and counteract proinflammatory signaling mediated by estrogen receptor beta. The anti-inflammatory action is mediated by progesterone receptor chaperones HSP90, FKBP51, and FKBP52. Drugs targeting these molecules, such as HSP90 inhibitors, can restore the lost anti-inflammatory effect of glucocorticoids and progesterone in chronic inflammatory and autoimmune diseases.
