Proteomics-based vaccine targets annotation and design of subunit and mRNA-based vaccines for Monkeypox virus (MPXV) against the recent outbreak

Summary: In this study, mRNA and multi-epitopes-based vaccines were designed against Monkeypox Virus (MPXV) using proteomics and structural vaccinology approaches. Ten potential vaccine targets were identified from the whole proteome of MPXV, and potential epitopes of these proteins were mapped using structural vaccinology approaches for B cell, cytotoxic T lymphocytes (CTL), and Helper T lymphocytes (HTL). A multi-epitope vaccine (MVC) and mRNA-based vaccines were constructed by joining the epitopes together. The designed MVC showed robust interaction with toll-like receptor 2 (TLR2) and efficient expression in E. Coli K12 strain, suggesting its potential for inducing an immune response against MPXV.
