Attacking the PI3K/Akt/mTOR signaling pathway for targeted therapeutic treatment in human cancer

Le Yu et al.

Summary: This article reviews the mechanisms leading to the hyperactivation of PI3K/Akt/mTOR signaling in cancer and summarizes the efforts in developing PI3K/Akt/mTOR inhibitors as monotherapy or combination therapy in different cancer settings. Targeting the PI3K/Akt/mTOR signaling pathway through the development of new agents, drug delivery systems, or combination regimens provides insights into possible future directions for targeted therapeutic regimen and effective patient stratification strategy to improve clinical outcomes for cancer patients with deregulated PI3K/Akt/mTOR signaling.

Amygdalin potentiates the anti-cancer effect of Sorafenib on Ehrlich ascites carcinoma and ameliorates the associated liver damage

Attia Ahmed Attia et al.

Summary: This study aimed to investigate the anti-tumor properties of amygdalin against Ehrlich ascites carcinoma (EAC) in mice and its protective properties against liver damage. The results showed that amygdalin reduced tumor burden, improved liver function parameters, and decreased oxidative stress levels in the liver. Furthermore, it was suggested that amygdalin may exert its effects through regulating the expression of antioxidant and angiogenesis-related genes.

Effect of amygdalin on MCF-7, MDA-MB-231 and T-47D breast cancer cells in the in vitro study

Aleksandra Zielinska et al.

Summary: This study evaluated the effects of amygdalin on three breast cancer cell lines and normal cells in vitro. The results showed that amygdalin can inhibit the proliferation and metabolic activity of breast cancer cells, and affect the transcription levels of apoptosis-related genes. The study suggests that breast cancer cells are more sensitive to amygdalin.

Molecular Imaging of Tumor Microenvironment to Assess the Effects of Locoregional Treatment for Hepatocellular Carcinoma

Quan Chen et al.

Summary: Liver cancer, particularly hepatocellular carcinoma (HCC), is a major cause of cancer-related deaths globally. The heterogeneity of HCC and complexities within the tumor microenvironment pose challenges to long-term treatment efficacy. Therefore, new approaches for noninvasively assessing treatment responses and aiding clinical decision-making are urgently needed.

Amygdalin Decreases Adhesion and Migration of MDA-MB-231 and MCF-7 Breast Cancer Cell Lines

Bashir Mosayyebi et al.

Summary: The study demonstrated that amygdalin has cytotoxic effects on breast cancer cell lines, while being non-toxic to human skin fibroblast cell line. Amygdalin treatment resulted in decreased adhesion of cancer cells to fibronectin and collagen I, as well as reduced cell migration, particularly in MDA-MB-231 cells. Integrin alpha and beta levels were differentially regulated by amygdalin in both cancer cell lines, suggesting a role in cancer cell adhesion and migration.

Summary: This study found that p53 haploinsufficiency facilitates mTOR signaling, promoting HCC tumorigenesis and lung metastasis. Inhibition of PI3K/Akt reduces mTOR activity, and rapamycin significantly reduces hepatocarcinogenesis triggered by Tsc1 loss and p53 haploinsufficiency.

Inflammation, It's Regulation and Antiphlogistic Effect of the Cyanogenic Glycoside Amygdalin

Daniela Figurova et al.

Summary: Inflammation accompanies any disease process in vascularized metazoans. Efforts have been made to develop selective drugs aimed at removing pain, fever, or swelling, but serious adverse side effects of such inhibitors have become apparent. Scientific research has explored new possibilities, including naturally available substances like amygdalin, but its toxicity at different doses and effects on the body's inflammatory response remain unclear.

Ahmed M. Mamdouh et al.

Summary: The study investigated the anti-HCC efficacy of amygdalin, metformin, and their combination compared to doxorubicin. Results showed that the combination therapy was more cytotoxic and had higher ability to induce apoptosis and arrest cell cycle. In vivo, doxorubicin was effective in treating HCC, while the combination therapy showed promise as a potential treatment approach with low side effects.

Summary: Pyroptosis, apoptosis and necroptosis are well-defined programmed cell death pathways, with growing evidence showing extensive crosstalk among them. PANoptosis, regulated by the PANoptosome complex, combines features of pyroptosis, apoptosis, and/or necroptosis. This new concept is critically involved in various diseases and may inform the development of new therapeutics.
