Applications and challenges of ultra-small particle size nanoparticles in tumor therapy

Summary: With the development of nanotechnology, nanomedicines are widely used in tumor therapy. However, biological barriers in the delivery of nanoparticles still limit their application in tumor therapy. In order to overcome this limitation, the use of ultra-small nanoparticles is proposed, which can facilitate deep tumor penetration and uniform drug distribution. This review systematically summarizes the latest progresses and advantages of ultra-small nanoparticles, as well as discusses the perspectives and challenges of their strategy for cancer treatment.

Summary: Recombinant anti-B7-H3xCD3 bispecific antibodies (biAbs) were prepared and MMP-2-sensitive S-biAb/dEGCG@NPs were constructed for efficient and systemic glioblastoma (GBM) elimination. S-biAb/dEGCG@NPs showed enhanced intracranial accumulation compared to biAb/dEGCG@NPs, biAb/dEGCG complexes, and free biAbs. Furthermore, S-biAb/dEGCG@NPs induced GBM elimination by boosting the ferroptosis effect and enhancing immune checkpoint blockade (ICB) immunotherapy, making them promising antibody nanocarriers for enhanced cancer therapy.

Summary: Glioblastoma (GBM) is an aggressive tumor with high heterogeneity. The accumulation of fatty acid metabolites is associated with low survival rates in patients with high-grade gliomas. The diversity of brain lipids, especially polyunsaturated fatty acids (PUFAs), and the involvement of FA transport proteins and FA translocases are crucial for PUFAs transportation through the blood-brain barrier and delivery to brain cells. Among them, the CD36 FA translocase plays a role in the uptake of long-chain FAs and oxidized lipoproteins. Furthermore, the interaction between CD36 and thrombospondin-1 (TSP-1) has an important role in glioma cell invasion and angiogenesis in GBM. The interconnection and interrelationship between FA, CD36, CD47, and their ligand TSP-1 have been highlighted in recent research to exert a cumulative effect on the modulation of the GBM molecular network.

A reactive oxygen species-replenishing coordination polymer nanomedicine disrupts redox homeostasis and induces concurrent apoptosis-ferroptosis for combinational cancer therapy

Summary: We developed a doxorubicin-incorporated iron coordination polymer nanoparticle (PCFD) for efficient chemo-chemodynamic cancer therapy by using a cinnamaldehyde-based reactive oxygen species (ROS)-replenishing organic ligand. This ligand can release cinnamaldehyde in response to ROS, supplementing intracellular H2O2 and depleting glutathione (GSH) through a thiol-Michael addition reaction. Together with doxorubicin-induced ROS upregulation and Fe3+-enabled GSH depletion, it facilitates efficient doxorubicin release and enhanced Fenton reaction, leading to redox dyshomeostasis and cancer cell death through concurrent apoptosis-ferroptosis. In vitro and in vivo studies show that the ROS-replenishing PCFD exhibits a better anticancer effect compared to the ROS-consuming counterpart. This study provides a facile and straightforward strategy for designing ROS amplifying nanoplatforms for cancer treatment.
