Lipid nanoparticles-loaded with toxin mRNA represents a new strategy for the treatment of solid tumors

Summary: Cancer therapy has advanced greatly in the past decade, providing new strategies to inhibit cancer cell growth using immune modulation and gene therapy. Recent advances in mRNA delivery have shown the potential of mRNA-based vaccines and immune-modulators for cancer therapeutics. In our study, we demonstrated that local administration of LNPs entrapping a modified mRNA encoding a bacterial toxin induced significant anti-tumor effects and improved overall survival of treated mice. We propose mmRNA-loaded LNPs as a new class of anti-tumoral, toxin-based therapy.

The mechanism of action of N-acetylcysteine (NAC): The emerging role of H2S and sulfane sulfur species

Summary: Originally used as a mucolytic, N-acetylcysteine (NAC) is now a standard treatment for paracetamol poisoning and a widely used antioxidant. In addition to its traditional mechanisms, a newly discovered mechanism involves the conversion of NAC into hydrogen sulfide and sulfane sulfur species, which may explain many of its effects.

Summary: The kinetic characteristics of the interaction of N-acetylcysteine with reactive oxygen species, peroxyl radicals, and hydrogen peroxide were studied, revealing similarities to glutathione. Isothermal calorimetry was used to study the heat release kinetics of the interaction of GSH and ASH with H2O2 for the first time. The results suggest relative autonomy of the S-H and S-S bonds in thiols and disulfides, and similar radical initiation rates between ASH and GSH in chain propagation reactions.
