Leucine retention in lysosomes is regulated by starvation

Urmi Bandyopadhyay et al.

Summary: This study uncovers the lysosomal retention of leucine under starvation conditions and reveals that stored leucine is utilized in protein synthesis, which is regulated by specific proteins.

The H+-ATPase (V-ATPase): from proton pump to signaling complex in health and disease

Amity F. Eaton et al.

Summary: The primary function of H+-ATPase is to create an electrochemical proton gradient across eukaryotic cell membranes to energize cellular processes. It acidifies intracellular vesicles and organelles for essential cell biological events. Additionally, V-ATPases play central roles in normal and pathophysiological processes that impact human health, influencing areas such as cancer, neurodegenerative diseases, and sensory perception.

Growth Inhibition by Amino Acids in Saccharomyces cerevisiae

Stephanie J. Ruiz et al.

Summary: Amino acids are essential for metabolism but can be toxic at high levels intracellularly. Downregulation of amino acid transporters in Saccharomyces cerevisiae is a mechanism to avoid toxicity. Overexpression of certain amino acid transporters can induce growth defects, and various amino acids are growth inhibitory to yeast cells when transported into the cell at high levels.

Growth-dependent signals drive an increase in early G1 cyclin concentration to link cell cycle entry with cell growth

Robert A. Sommer et al.

Summary: The entry into the cell cycle is initiated by the cyclin Cln3, which drives an increase in cell cycle entry by inactivating Whi5. The concentration changes of Whi5 do not play a major role in controlling the cell cycle entry process, instead, cell growth triggers the entry by increasing the concentration of Cln3. Accumulation of Cln3 is dependent upon homologs of mammalian SGK kinases, suggesting a crucial link between cell growth and the cell cycle.

The metabolic growth limitations of petite cells lacking the mitochondrial genome

Jakob Vowinckel et al.

Summary: The study reveals that petite cells suffer from an insufficient capacity to synthesize glutamate, glutamine, leucine and arginine, negatively impacting their growth rate. Fast growth overcomes these amino acid deficiencies by alleviating a perturbation in mitochondrial iron metabolism and restoring a defect in the mitochondrial tricarboxylic acid cycle.
