A pH-responsive CaO2@ZIF-67 system endows a scaffold with chemodynamic therapy properties

Guowen Qian et al.

Summary: This study constructs a pH-responsive CaO2@ZIF-67 system by in-situ growing ZIF-67 on the surface of CaO2, and combines it with PLLA to prepare a PLLA/CaO2@ZIF-67 scaffold using selective laser sintering technology. The results show that the scaffold can sustainably produce H2O2 under acidic pH and exhibit an antitumor ratio of 84.64% against MG-63 cells. Additionally, the scaffold promotes the proliferation and osteogenic differentiation of mouse bone marrow mesenchymal stem cells through the release of calcium ions. This PLLA/CaO2@ZIF-67 scaffold with chemodynamic antitumor and osteogenic effects has great potential for treating tumorous bone defects.

Non-spherical Polymeric Nanocarriers for Therapeutics: The Effect of Shape on Biological Systems and Drug Delivery Properties

Prescillia Lagarrigue et al.

Summary: This review emphasizes the importance of particle shape in the design of polymeric nanocarriers for drug delivery systems, in addition to their size, surface chemistry, density, and rigidity. The current manufacturing methods used to obtain non-spherical polymeric nanocarriers such as filomicelles or nanoworms, nanorods and nanodisks are firstly described. Then, the interactions of these nanocarriers with biological barriers are presented, including how shape influences nanoparticle clearance, biodistribution, and targeting. Finally, their drug delivery properties and therapeutic efficacy, both in vitro and in vivo, are discussed and compared with their spherical counterparts.

Magnetotactic Bacteria-Based Drug-Loaded Micromotors for Highly Efficient Magnetic and Biological Double-Targeted Tumor Therapy

Bo Wang et al.

Summary: Bacteria-mediated cancer therapy has gained attention, but using magnetotactic bacteria for therapy has not been reported. In this study, a photosensitizer was incorporated into a bacterial strain M. magneticum AMB-1, creating AMB-1/Ce6 micromotors with high drug payload and bacterial activities. It was found that AMB-1 not only acts as a drug carrier but also inhibits tumor growth. Under laser irradiation, micromotors efficiently killed cancer cells by generating reactive oxygen species. They could target hypoxic and normoxic regions through active swimming and magnetic field guidance, showing high tumor-homing ability. The micromotors significantly inhibited mouse tumor growth through biotherapy and phototoxicity. This strategy may be a promising platform for treating solid tumors.

Magnetic Microcarriers with Accurate Localization and Proliferation of Mesenchymal Stem Cell for Cartilage Defects Repairing

Mengjiao Ma et al.

Summary: In this study, a magnetically responsive composite microcarrier was developed for tissue engineering applications. The microcarrier exhibited paramagnetic properties and showed suitable characteristics for cell growth and biocompatibility. Culturing bone marrow mesenchymal stem cells (BMSCs) on the magnetic microcarriers, and applying a static magnetic field (SMF) resulted in enhanced cell proliferation. Implanting the magnetic microcarriers loaded with BMSCs into a rat model of cartilage defect and adding a magnet to the operative side led to improved cartilage regeneration and repair. The study suggests that magnetic composite microcarriers hold promise as tissue-engineered scaffolds for cartilage regeneration and repair.
