Beneficial effects of trehalose and gentiobiose on human sperm cryopreservation

Summary: Sperm cryopreservation is important for maintaining genetic diversity in chicken breeds, but many sperm lose their viability and motility during the freezing/thawing process. Cell membranes, especially the lipid composition, play a crucial role in cryoresistance. By studying the lipid composition of spermatozoa membranes in two breeds of roosters, a cryoprotective diluent with a concentration of 9.5 mM trehalose was developed. This diluent helps maintain a high level of cell lipids and the kinetic ability of frozen/thawed spermatozoa at 52.4%.

The In Vitro Evaluation of Rooster Semen Pellets Frozen with Dimethylacetamide

Summary: This study aims to evaluate the impact of different concentrations (3%, 6%, or 9%) of dimethylacetamide (DMA) as a cryoprotectant on post-thawed sperm quality, motility, antioxidant-biomarkers, and the expression of anti-freeze related genes. The results showed that 3% DMA group maintained higher post-thawed sperm quality compared to the other tested groups, with improvements in motility, viability, and antioxidant enzyme activity. Additionally, certain anti-freeze related genes were upregulated in the 3% DMA group.

Summary: Ex situ preservation is an important method for preserving chickens, and currently cryopreservation of semen is the only method available for gamete preservation. It is hoped that this review will serve as a reference for future investigations on poultry semen cryopreservation technology.

Summary: This study aimed to investigate the effects of various methods of preparing rooster semen on its quality characteristics and evaluate the change in the composition of spermatozoon cytosol during equilibration and short-term storage. The study found that filtered semen had higher progressive motility and chromatin integrity compared to centrifuged semen. Additionally, the study identified inositol as the main component in the composition of spermatozoa cytosol and suggested its role as the main antioxidant. The introduction of inositol into synthetic diluents, including cryoprotective ones, was deemed biologically justified.
