Comparing the efficacy in reducing brain injury of different neuroprotective agents following neonatal hypoxia-ischemia in newborn rats: a multi-drug randomized controlled screening trial

Summary: Intrapartum hypoxia-ischemia leading to neonatal encephalopathy is a major cause of neonatal mortality and morbidity worldwide, particularly in low- and middle-income countries. Therapeutic hypothermia is currently the only effective treatment available in high-income countries, but its effectiveness is limited in LMIC. In this study, we compared the effects of 25 potential neuroprotective drug candidates on neonatal hypoxic-ischemic brain injury in rats. We found that caffeine, Sonic Hedgehog Agonist (SAG), and Allopurinol had the strongest treatment effects, followed by Melatonin, Clemastine, ss-Hydroxybutyrate, Omegaven, and Iodide. These alternative therapies may hold promise for the treatment of hypoxic-ischemic brain injury in LMIC.

Drug repurposing: Clemastine fumarate and neurodegeneration

Summary: Neurodegenerative diseases are a serious problem in older people, causing motor and cognitive defects and lowering their quality of life, necessitating effective treatment. Drug repurposing, the exploration of existing drugs for new therapeutic purposes, has become a wise and popular strategy to provide new treatment options. Clemastine fumarate, a promising drug for remyelination and autophagy enhancement, has been identified through screening. This review summarizes the potential mechanisms of clemastine fumarate in current literature and aims to find worthwhile research directions for neurodegeneration diseases by distinguishing the independent or synergistic effects between clemastine and fumarate.

Hypoxic oligodendrocyte precursor cell-derived VEGFA is associated with blood-brain barrier impairment

Summary: Cerebral small vessel disease is characterized by decreased cerebral blood flow and blood-brain barrier impairments, leading to the development of white matter lesions. Cerebral hypoperfusion induces hypoxia and affects the signaling between oligodendrocyte precursor cells and endothelial cells, resulting in blood-brain barrier dysfunction. In this study, a mouse model of cerebral hypoperfusion was used to investigate the role of hypoxia-induced VEGFA expression in the development of white matter lesions. The results suggest that VEGFA upregulation in hypoxic oligodendrocyte precursor cells mediates blood-brain barrier permeability, which is associated with increased VEGFA plasma levels in cerebral small vessel disease patients.

Increased Systemic Immune-Inflammation Index Predicts Disease Severity and Functional Outcome in Acute Ischemic Stroke Patients

Summary: This study aimed to investigate the relationship between systemic immune-inflammation index (SII) and system inflammation response index (SIRI) and severity of stroke, and to analyze their prognostic value in acute ischemic stroke (AIS) patients. The results showed that higher SII and SIRI indexes were associated with greater risk of stroke severity, while SII could be useful for predicting adverse clinical outcomes after AIS.

Clemastine Induces an Impairment in Developmental Myelination
