Ligands for Abasic Site-containing DNA and their Use as Fluorescent Probes

Summary: Apurinic and apyrimidinic sites are residues of duplex DNA with missing bases, which can be used for DNA repair and provide binding sites for guest molecules. Ligands that selectively bind to abasic sites can be used for analytical and therapeutic purposes. This review provides an overview of different classes of ligands for abasic site-containing DNA, including covalently and non-covalently binding substrates. It also summarizes reports on the fluorescent response of ligands upon binding to AP-DNA and presents a perspective for future development of fluorescent probes for AP-DNA.

Phototunable Cell Killing by Photochromic Diarylethene of Thiazoyl and Thienyl Derivatives

Summary: We have developed a novel technique for cell killing using diarylethene molecules as photo-isomerizing-molecular switches. These molecules are delivered to the cell nucleus, where a UV light irradiation switches them into a closed form that induces DNA intercalation. Subsequent blue light irradiation triggers cell killing, leading to DNA double-strand breaks. The unique photo-controllable action of these molecules provides a basis for a new phototherapy scheme.

Formation of Environmentally Persistent Free Radicals from the Irradiation of Polycyclic Aromatic Hydrocarbons

Summary: Polycyclic aromatic hydrocarbons (PAHs) provide a complex matrix for environmentally persistent free radicals (EPFRs) to stabilize in particulate matter and cause cardiopulmonary diseases. This study investigated the formation and transformation of EPFRs from four PAHs upon photochemical and aqueous-phase aging. The results showed the increase in EPFR concentrations and their impact on the environment due to atmospheric aging.

Pure DNA scaffolded drug delivery systems for cancer therapy

Research opportunities in the field are speculated to pave the way for future research direction.

Chiral rhodium(III)-azobenzene complexes as photoswitchable DNA molecular locks

Summary: Chiral rhodium(III)-azobenzene complexes capable of intercalating into DNA were developed. Upon light exposure, the azobenzene moiety of the metal complexes can undergo photoisomerization from trans-form to cis-form, thereby stabilizing the DNA double-helix and modulating DNA transcription. This study presents the first example of metal-based photoswitchable DNA molecular locks.

Machine Learning Aids Classification and Discrimination of Noncanonical DNA Folding Motifs by an Arrayed Host:Guest Sensing System

Summary: An arrayed host:guest fluorescence sensor system can accurately discriminate and classify multiple different noncanonical DNA structures through selective molecular recognition. The sensor is highly selective and can distinguish between folds with subtle differences, such as native G-quadruplexes and those with bulges or vacancies. By forming heteroternary complexes with DNA strands, the host mediates the interaction between DNA and dye to modulate emission, and machine learning algorithms enable high-fidelity prediction of the folding state of unknown DNA strands.
