3-Amino-Substituted Analogues of Fusidic Acid as Membrane-Active Antibacterial Compounds

Elena V. Salimova et al.

Summary: The study aimed to investigate the antimicrobial effect of FA amines and their potential biological targets. It was found that pyrazinecarboxamide-substituted analogues of FA exhibited high antibacterial activity against MRSA. Furthermore, FA derivatives containing spermine and spermidine were effective against Gram-negative bacteria and had fungicidal effects. The study also demonstrated that these compounds could interact with cell membranes and disrupt lipids, and spermine-based compounds could form ion-permeable pores. Molecular docking suggested that the inhibition of the protein synthesis elongation factor EF-G played a key role in the antibacterial effect of these compounds.

BKCa Activator NS1619 Improves the Structure and Function of Skeletal Muscle Mitochondria in Duchenne Dystrophy

Mikhail Dubinin et al.

Summary: This study shows that mitochondrial dysfunction and potassium imbalance play a role in the development of Duchenne muscular dystrophy. Activating the mitoBK(Ca) channel can improve mitochondrial function and potassium homeostasis, leading to a reduction in muscle degeneration and fibrosis.

Effect of Modified Levopimaric Acid Diene Adducts on Mitochondrial and Liposome Membranes

Mikhail Dubinin et al.

Summary: This paper demonstrates the membranotropic effect of modified levopimaric acid diene adducts on liver mitochondria and lecithin liposomes. The derivatives reduced the efficiency of oxidative phosphorylation in mitochondria and affected the membrane potential in organelle energization. The compounds also exhibited surface-active properties and induced nonspecific permeabilization of mitochondrial membranes and swelling of the organelles.

New quaternized pyridinium derivatives of betulin: Synthesis and evaluation of membranotropic properties on liposomes, pro- and eukaryotic cells, and isolated mitochondria

Mikhail Dubinin et al.

Summary: The study synthesized pyridinium derivatives of betulin, showing their effects on artificial membrane systems and cytotoxicity in cell models. The presence of methyl groups in the compounds influenced their ability to permeabilize membranes and affect mitochondrial function. The study highlights the dependence of membranotropic and biological actions on the structure and lipophilicity of the derivatives.

Repurposing primaquine as a polyamine conjugate to become an antibiotic adjuvant

A. Norrie Pearce et al.

Summary: Evaluation of succinylprimaquine-polyamine conjugates identified two derivatives with intrinsic antimicrobial activity against Staphylococcus aureus and Cryptococcus neoformans. In the presence of doxycycline, these compounds also showed potent antibiotic adjuvant properties against Pseudomonas aeruginosa. Further studies explored the structure-activity relationship to enhance antimicrobial properties against P. aeruginosa.
