A panel of visual bacterial biosensors for the rapid detection of genotoxic and oxidative damage: A proof of concept study

Summary: The emergence of new compounds in the past decade requires a high-throughput screening method for toxicity assay. In this proof-of-concept study, stress-responsive whole-cell biosensors were used to evaluate the damages induced by toxic chemicals. Nine stress-responsive promoters were selected to assemble a set of blue indigoidine-based biosensors. Dose-dependent increase of visible blue signal was observed in response to mutagens, showing the potential of these biosensors for detecting DNA damage and oxidative stress.

Enteric pathogen reduction targets for onsite non-potable water systems: A critical evaluation

Summary: In this work, the authors compare and synthesize the log10-reduction targets (LRTs) for pathogens in onsite non-potable water systems (ONWS) to inform the selection of pathogen LRTs. The results show that there were minor differences in LRTs for human enteric viruses and parasitic protozoa between 2017 and 2021 efforts, despite differences in pathogen characterization methods. The greatest differences were observed in stormwater due to the use of municipal wastewater data and different reference pathogens.

Development of silver nanoparticles and aptamer conjugated biosensor for rapid detection of E. coli in a water sample

Summary: A simple, rapid, and sensitive electrochemical biosensor based on a screen-printed carbon electrode was developed for onsite detection of E. coli. The effect of aptamer conjugation and PBS buffer solution on the colloidal stability of AgNPs was analyzed. The biosensor showed good sensitivity with a limit of detection of 150 CFU/ml and selectivity for E. coli compared to other bacteria. It also exhibited good reproducibility, stability, and high recoveries during sensitivity validation.

A Colorimetric Aptasensor for Ochratoxin A Detection Based on Tetramethylrhodamine Charge Effect-Assisted Silver Enhancement

Summary: A novel colorimetric aptasensor was developed to detect ochratoxin A (OTA) by utilizing charge effect-assisted silver enhancement. Gold nanoparticles (AuNPs) were used as carriers of aptamers tagged with a positively charged tetramethylrhodamine (TAMRA), which catalyzed silver reduction and deposition. The TAMRA attracted and retained the silver lactate around the AuNPs due to the attraction between positive and negative charges, increasing the chance of AuNP-catalyzed silver reduction. The charge effect-assisted silver enhancement was confirmed by tagging aptamers of different base pair lengths with TAMRA. The as-prepared OTA aptasensor exhibited a working range of 1 x 10(2)-1 x 10(6) pg mL(-1) and a detection limit of 28.18 pg mL(-1). It was successfully applied to determine OTA in actual samples with satisfactory results.
