DNA walker for signal amplification in living cells

Summary: The rapid development of DNA nanotechnology allows for the programmable operation of advanced DNA walkers, which mimic the function of protein motors in living organisms. These walkers require four main components for construction, including motor, track, energy, and cargo, and can autonomously perform specific functions following a predesigned trajectory with nanometer precision. Recent research has focused on improving the progressivity, speed, and directionality of DNA walkers, and new DNA nano techniques have shown powerful signal amplification during their operation. This review covers the breakthrough design and principle of walking systems and highlights the recent application of DNA walkers for signal amplification in living cells. The engineered DNA walkers have great potential for diverse applications, such as imaging intracellular molecules, modulating cellular functions, and facilitating drug delivery.

Electrochemical determination of Pb2+based on DNAzyme-triggered rolling circle amplification and DNA-templated silver nanoclusters amplification strategy

Summary: An electrochemical biosensor based on rolling circle amplification (RCA) was developed for the detection of lead ion (Pb2+). The biosensor utilized Pb2+-dependent DNAzyme and DNA-templated silver nanoclusters (DNA-AgNCs) for amplification, and showed high sensitivity and selectivity for Pb2+ analysis in the range of 1 pM-10 μM with a detection limit of 0.3 pM. This strategy has potential applications in real water samples and environment analysis.

Engineering DNAzyme strategies for fluorescent detection of lead ions based on RNA cleavage-propelled signal amplification

Summary: Based on the high recognition ability and flexible programmability of GR5 DNAzyme, two fluorescent biosensors were developed for amplified detection of Pb2+ using Ti3C2TX MXenes and 2-aminopurine (2-AP). The sensors demonstrated simple procedures, high sensitivity, and good specificity, making them suitable for on-site monitoring of lead pollution in water samples.

Development of regional water quality criteria of lead for protecting aquatic organism in Taihu Lake, China

Summary: Lead is a widely distributed heavy metal with priority control in aquatic systems, and its toxicity to aquatic organisms is influenced by water quality parameters. This study investigated the acute toxicity of lead to aquatic organisms in Taihu Lake, deriving corresponding regional water quality criteria based on different water hardness levels. The results showed that lead toxicity to aquatic organisms increased with water hardness, with the highest toxicity observed at a water hardness of 50 mg/L (expressed as CaCO3).
