Microfluidics for COVID-19: From Current Work to Future Perspective

Summary: The spread of COVID-19 has had a significant impact on public health and the economy, requiring the urgent development of rapid, convenient, and cost-effective point-of-care testing (POCT) technologies to diagnose and control the transmission of the disease. Microfluidics, with its advantages of miniaturization, high throughput, small sample requirements, and low consumption, plays a critical role in the development of POCT devices. This review introduces common microfluidic-based POCT devices on the market, such as paper-based, centrifugal, optical, and digital platforms, and summarizes various microfluidic-based assays for diagnosing COVID-19, including immunoassays (such as ELISA) and molecular assays (such as PCR). The challenges and future perspectives of microfluidic device design and development are also discussed.

Hundred-micron droplet ejection by focused ultrasound for genomic applications

Summary: Droplet-based microfluidic (DMF) technology is widely used in genetic analysis. Acoustic droplet ejection (ADE) is a promising non-contact method for controlled droplet generation and nucleic acid manipulation in droplets. This study successfully achieved droplet ejection using a focused ultrasonic transducer and demonstrated the scalability of droplet size and the feasibility of multi-droplet ejections. Furthermore, it was shown that nucleic acid molecules could maintain their integrity under maximum power and longest duration of excitation in the ADE system.

On-Chip Nucleic Acid Purification Followed by ddPCR for SARS-CoV-2 Detection

Summary: We developed a microfluidic chip that combines nucleic acid purification and droplet-based digital polymerase chain reaction (ddPCR) modules for infectious virus diagnosis. The chip achieved a "sample-in, result-out" process by utilizing magnetic beads and oil-enclosed droplets. The chip demonstrated good uniformity, adjustable droplet diameters, and controllable flow rates, and it was able to accurately detect the concentration of nucleic acids from the SARS-CoV-2 virus. The chip's on-chip purification and detection abilities make it a valuable tool for point-of-care testing.

OsciDrop: A Versatile Deterministic Droplet Generator

Precise monodisperse droplet generation by pressure-driven microfluidic flows

Summary: A mathematical model describing the nonlinear relationship between droplet generation in a T-junction microdroplet generator is established, showing that the droplet length varies nonlinearly with the driving-pressure ratio under low capillary numbers. Experimental testing of the droplet length and driving-pressure ratio relationship is done for different T-junction geometries. A closed-loop control droplet microfluidic system is demonstrated to more accurately control droplet size, with the relative standard deviation of droplet size under different flow conditions being less than 2%.

Comparison of formation of bubbles and droplets in step-emulsification microfluidic devices

By changing the placement of the microdevice, the effects of buoyancy and gravity on the generation of bubbles and droplets are introduced, and the feedback mechanism of the bubble layer and the effect of droplet accumulation on the emulsification process are clarified.
