Applications, Challenges, and Outlook for PBPK Modeling and Simulation: A Regulatory, Industrial and Academic Perspective

Wen Lin et al.

Summary: This review discusses the applications of physiologically based pharmacokinetic (PBPK) models and physiologically based biopharmaceutics models (PBBMs) in drug development. It proposes practical considerations for constructing and developing base PBPK models and summarizes the current status, challenges, and future perspectives in PBPK evaluation for various scenarios such as drug-drug interactions, pediatric patients, organ impairment, maternal-fetal drug disposition, and pH-mediated drug interactions.

Physiologically-based toxicokinetic modeling of human dermal exposure to diethyl phthalate: Application to health risk assessment

Man Hu et al.

Summary: This study developed a dermal physiologically based toxicokinetics (PBTK) model for diethyl phthalate (DEP) exposure via personal care products (PCPs) and indoor air in humans. The results showed that dermal exposure to DEP poses a greater risk to human health compared with oral exposure.

Predicting the systemic exposure and lung concentration of nafamostat using physiologically-based pharmacokinetic modeling

Hyeon-Cheol Jeong et al.

Summary: This study predicted and evaluated the pharmacokinetic profiles of Nafamostat in a virtual healthy population using a PBPK model. The results suggested that the proposed dosing regimens for COVID-19 treatment could maintain the effective concentration for inhibiting coronavirus entry into plasma and lung tissue.

Physiologically Based Pharmacokinetic Modeling of Rosuvastatin to Predict Transporter-Mediated Drug-Drug Interactions

Nina Hanke et al.

Summary: A PBPK model of the drug rosuvastatin was built for investigating its pharmacokinetics and transporter-mediated drug-drug interactions. By incorporating extensive clinical data and DDI studies, the model accurately described rosuvastatin metabolism and successfully predicted DDIs.
