PRMT4 promotes ferroptosis to aggravate doxorubicin-induced cardiomyopathy via inhibition of the Nrf2/GPX4 pathway

Yilong Wang et al.

Summary: PRMT4 promotes ferroptosis in DOX-induced cardiomyocyte ferroptosis by interacting with Nrf2 and inhibiting its nuclear translocation, which subsequently suppresses the transcription of GPX4. Activation of Nrf2 or administration of Fer-1 abolishes the detrimental role of PRMT4 in DOX-induced cardiomyocyte ferroptosis.

Ethyl carbamate triggers ferroptosis in liver through inhibiting GSH synthesis and suppressing Nrf2 activation

Yang Xu et al.

Summary: This study found that humans are exposed to ethyl carbamate (EC) through consumption of fermented food and beverages. EC triggers ferroptosis in liver cells, causing cell death. GSH depletion is the main cause of ferroptosis under EC exposure. EC blocks the activation of Nrf2, leading to the occurrence of ferroptosis.

Alpha-lipoic acid treatment improves adverse cardiac remodelling in the diabetic heart - The role of cardiac hydrogen sulfide-synthesizing enzymes

George J. Dugbartey et al.

Summary: Alpha-lipoic acid (ALA) is a licensed drug for the treatment of diabetic neuropathy and diabetic cardiomyopathy (DCM) in type 2 diabetes mellitus (T2DM). This study shows that ALA preserves myocardial integrity in T2DM by upregulating cardiac hydrogen sulfide (H2S)-synthesizing enzymes and increasing plasma H2S level.

Ferroptosis: molecular mechanisms and health implications

Daolin Tang et al.

Summary: Cell death can be executed through different subroutines, such as ferroptosis which is an iron-dependent form of non-apoptotic cell death. Ferroptosis can occur through two major pathways, driven by redox imbalance and abnormal expression of redox-active enzymes. The process is precisely regulated at multiple levels, with the transcription factor NFE2L2 playing a central role in anti-ferroptotic defense.
