Preclinical Evaluation of TB/FLU-04L-An Intranasal Influenza Vector-Based Boost Vaccine against Tuberculosis

Summary: Tuberculosis is a global health threat, and a new boost tuberculosis vaccine called TB/FLU-04L was developed. This vaccine uses an attenuated influenza A virus as a vector and encodes two mycobacterium antigens. Intranasal immunization with TB/FLU-04L induced a specific Th1 immune response and provided a level of protection comparable to BCG. Additionally, it significantly enhanced the protective effect of BCG in a prime-boost scheme.

Clinical manifestations and immune response to tuberculosis

Despite a large number of individuals being carriers of the tuberculosis bacillus, only a third of them develop active disease. Understanding the relationship between clinical manifestations of tuberculosis and the immune response can provide insights into the pathophysiological and immunological mechanisms of the disease.

Vaccines against Tuberculosis: Where Are We Now?

Summary: Tuberculosis (TB) is a major cause of death, killing over 30,000 people per week and surpassing other infectious diseases like AIDS and malaria. TB treatment is affected by factors such as ineffective drugs, lack of advanced vaccines, misdiagnosis, improper treatment, and social stigma. Different vaccine strategies have been employed, including protein subunit vaccines, viral vector vaccines, whole-cell inactivation vaccines, and recombinant BCG vaccines. Currently, there are approximately 19 vaccine candidates in clinical trials. Advanced vaccines that generate heterologous immune responses are needed to enhance long-lasting immunity against both drug-sensitive and drug-resistant TB.

Integer time series models for tuberculosis in Africa

Immune Regulatory Effect of Osteopontin Gene Therapy in a Murine Model of Multidrug Resistant Pulmonary Tuberculosis

Transient, inhaled gene therapy with gamma interferon mitigates pathology induced by host response in a mouse model of tuberculosis

Summary: Transient transfection with gamma interferon (IFN-??) on the respiratory mucosa of mice infected with Mycobacterium tuberculosis (Mtb) shows promising benefits in disease therapy. Dry powder inhalation (DPI) as a stand-alone or as an adjunct to oral anti-tuberculosis (TB) chemotherapy effectively mitigates the organ pathology and histopathology in infected mice. However, DPI alone does not result in a significant reduction in Mtb colony forming units (CFU). The frequency of DPI administration (one or three doses per week) does not significantly affect lung CFU, but a lower frequency (four doses) shows better results in mitigating lung pathology.
