Benefit of Adjuvant Mesenchymal Stem Cell Transplantation to Critical-Sized Peripheral Nerve Defect Repair: A Systematic Review and Meta-Analysis of Preclinical Studies

Summary: Critically sized nerve defects require reconstruction for lifelong disabilities. Local application of mesenchymal stem cells (MSCs) is considered promising in enhancing peripheral nerve regeneration. A systematic review and meta-analysis were conducted to evaluate the effects of MSCs in preclinical studies. The results showed that co-transplantation of MSCs increased functional index, nerve conduction velocity recovery, muscle atrophy, and axon regeneration in rats with critically sized defects. These findings demonstrate the potential clinical benefits of MSCs in peripheral nerve reconstruction.

Neural guidance channels (NGCs) and hydrogel-based drug delivery systems have been developed to address the challenges of autologous nerve grafting. Alginate, a naturally occurring polysaccharide, has shown promising results in the repair of peripheral nervous system injuries.

The effect of poly(lactic-co-glycolic acid) conduit loading insulin-like growth factor 1 modified by a collagen-binding domain on peripheral nerve injury in rats

Summary: This study designed and tested a collagen-coated poly(lactic-co-glycolic acid) conduit loaded with CBD-IGF-1 for nerve regeneration. The conduit showed a rough surface and good hydrophilicity, and CBD-IGF-1 could be released slowly. In a rat sciatic nerve injury model, the group treated with the conduit showed the best sciatic functional index and electrophysiological parameters, along with better pathological examination results compared to the other groups.
