Optogenetic approaches in biotechnology and biomaterials

Vasily V. Reshetnikov et al.

Summary: Advances in genetic engineering and optical technologies have expanded the possibilities of optogenetics in recent years. However, its application in industries such as biotechnology and biomaterials production is still limited, as specific optogenetic systems need to be engineered for each practical task. This review discusses the latest developments in using optogenetic tools in biofuel and valuable chemical production, biomedical and polymer material synthesis, and plant agrobiology. It also provides a comprehensive analysis of light-controlled and smart biomaterials' properties and industrial applicability, outlining the prospects for future use of optogenetics in the bioindustry.

Robust and tunable signal processing in mammalian cells via engineered covalent modification cycles

Ross D. Jones et al.

Summary: Phosphorylation networks play a crucial role in complex cellular decision making. This study engineers synthetic phosphorylation devices with feedback regulation in mammalian cells and demonstrates their tunable and robust control over cell behaviors. By using bacterial two-component signaling proteins, the authors develop synthetic phosphoregulation devices that can precisely regulate target gene expression and withstand perturbations in mammalian cells. The work lays the foundation for establishing tunable, precise, and robust control over cell behavior with synthetic signaling networks.

Synthetic gene networks recapitulate dynamic signal decoding and differential gene expression

Dirk Benzinger et al.

Summary: Cells employ dynamic signaling pathways to decode and process signals for gene regulation. Researchers have developed networked optogenetic pathways to achieve dynamic signal processing functions that mimic cellular information processing. The study shows that dynamic multiplexing significantly increases the information transmission capacity from signal to gene expression state and enables precise multidimensional regulation of metabolic pathways.

A genetic mammalian proportional-integral feedback control circuit for robust and precise gene regulation

Timothy Frei et al.

Summary: This study demonstrates the implementation of the antithetic integral feedback motif in mammalian cells, showing that it robustly maintains the expression of a synthetic transcription factor. By augmenting this motif with additional negative feedback, an improved regulatory strategy is achieved, which reduces the variance of the regulated transcription factor. It is suggested that the engineering of precise and robust perfect adaptation will have significant advances in industrial biotechnology and cell-based therapeutics.

Optogenetic Amplification Circuits for Light-Induced Metabolic Control

Evan M. Zhao et al.

Summary: OptoAMP is a new series of optogenetic circuits that amplify the transcriptional response to blue light up to 23-fold compared to the basal circuit (OptoEXP). These circuits show up to a 41-fold induction between dark and light conditions, efficient activation at light duty cycles as low as similar to 1%, and strong homogeneous light-induction in bioreactors of at least 5 L, with limited illumination at cell densities above 40 OD600.

An Automated Tabletop Continuous Culturing System with Multicolor Fluorescence Monitoring for Microbial Gene Expression and Long-Term Population Dynamics

Ue-Yu Pen et al.

Summary: Monitoring gene expression dynamics and population levels in a multispecies microbial community in real-time could provide insights into the impact of changing gene expression patterns on eco-evolutionary outcomes. A unique experimental platform with high dynamic range and temporal resolution was designed and validated for this purpose, showing its capability to capture gene expression dynamics and population dynamics in response to external perturbations and different nutrient conditions. The system demonstrated the ability to switch between competitive exclusion and long-term coexistence in a two species microbial community.
