A novel RHH family transcription factor aCcr1 and its viral homologs dictate cell cycle progression in archaea

Summary: Cell cycle regulation is crucial for all forms of life. A transcription factor called aCcr1 and its viral homologs control cell division in Sulfolobales. Over-expression of aCcr1 and its viral homologs leads to growth retardation and cell division failure, while its over-expression downregulates 17 genes, including cdvA. This study provides insights into the conserved mechanism of cell division regulation in archaeal cells and the manipulation of host cell cycle by viruses.

The Impact of Single-Stranded DNA-Binding Protein SSB and Putative SSB-Interacting Proteins on Genome Integrity in the Thermophilic Crenarchaeon Sulfolobus acidocaldarius

Summary: The study of DNA repair in hyperthermophiles can uncover the mechanisms of genome integrity maintenance systems in extreme conditions. Previous studies have shown that the single-stranded DNA-binding protein (SSB) from Sulfolobus is involved in maintaining genome integrity, including mutation avoidance, homologous recombination, and repair of helix-distorting DNA lesions. In this study, the mutant phenotypes of the SSB-deleted strain Delta ssb in S. acidocaldarius were characterized, revealing an increase in mutation rate and a defect in homologous recombination. This study expands our understanding of the impact of SSB on genome integrity and identifies key proteins involved in repair mechanisms in hyperthermophilic archaea.

A Well-Conserved Archaeal B-Family Polymerase Functions as an Extender in Translesion Synthesis

Summary: In this work, we discovered that Sulfolobus islandicus Dpo2, a B-family DNA polymerase once predicted to be inactive, is a functional DNA polymerase involved in translesion synthesis. S. islandicus Dpo2 belongs to a large group of B-family DNA polymerases (PolB2) that exists in many archaea and some bacteria, and they carry variations in conserved amino acids in the functional domains responsible for polymerization and proofreading. Contrary to predictions, this prokaryotic B-family DNA polymerase not only replicates undamaged DNA accurately, but also efficiently extends mismatched and DNA lesion-containing substrates. Based on our findings, we propose that this enzyme functions as an extender polymerase, representing the first prokaryotic enzyme of this type. Our data also suggests that this PolB2 enzyme serves as a functional counterpart of the eukaryotic DNA polymerase Pol zeta, which is dedicated to DNA damage repair.

A Novel Family of Winged-Helix Single-Stranded DNA-Binding Proteins from Archaea

Summary: This study reports a novel winged helix single-stranded DNA binding protein family called Sul7s, which exhibits an approximately 15-fold higher affinity for single-stranded DNA than for double-stranded DNA in vitro. The solution structure of Sul7s, determined by NMR, reveals a winged helix-turn-helix fold and interacts with single-stranded DNA through a large positively charged binding surface.

The production of EVs in bacteria is mediated by the ESCRT machinery, with a prime role of ESCRT-III-1 and ESCRT-III-2 proteins in EV budding. EVs have deep evolutionary roots and play a crucial role in horizontal gene transfer and nutrient cycling in extreme environments.
