Urate Transporter 1 Can Be a Therapeutic Target Molecule for Chronic Kidney Disease and Diabetic Kidney Disease: A Retrospective Longitudinal Study

Hidekatsu Yanai et al.

Summary: Chronic kidney disease (CKD) is a major global health problem, and there are currently no curative drug treatments available. Hyperuricemia is a risk factor for CKD, but the evidence on the effects of uric acid (UA)-lowering treatments on CKD progression is limited. Dotinurad, a selective urate reabsorption inhibitor, was found to be effective in reducing serum UA levels and improving metabolic parameters in CKD patients with hyperuricemia. Furthermore, dotinurad selectively targets URAT1, which may be beneficial for the pathology of CKD.

Direct cardio-protection of Dapagliflozin against obesity-related cardiomyopathy via NHE1/MAPK signaling

Ke Lin et al.

Summary: This study investigated the protective effect of Dapa against obesity-related cardiomyopathy caused by a high fat diet. The results showed that Dapa can reduce cardiomyocyte hypertrophy, fibrosis, and apoptosis, and alleviate cardiac dysfunction and remodeling caused by obesity. This protective effect is achieved through the inhibition of the MAPK/AP-1 pathway and improvement of cardiac inflammation. These findings highlight the potential use of Dapa in the prevention of obesity-related cardiac dysfunction.

Summary: This study demonstrates the direct hypothermic effect of T1AM on cardiomyocytes and its upregulation of BNP expression through the activation of the MEK/ERK pathway. These findings provide new insights into the cellular actions of T1AM in patients with severe heart failure.

Xanthine oxidase inhibition attenuates doxorubicin-induced cardiotoxicity in mice

Yoshiro Tanaka et al.

Summary: Accumulating evidence suggests that high serum uric acid is associated with left ventricular dysfunction, and xanthine oxidase activation plays a critical role in this process. In a study on doxorubicin-induced cardiotoxicity, it was found that cardiac tissue xanthine oxidase activation led to impaired left ventricular function, which was attenuated by xanthine oxidase inhibitors through reduction of oxidative stress and modulation of purine metabolism.

From Systemic Inflammation to Myocardial Fibrosis The Heart Failure With Preserved Ejection Fraction Paradigm Revisited

Walter J. Paulus et al.

Summary: According to the comorbidity-inflammation paradigm, comorbidities, especially metabolic ones, are believed to drive the development and severity of heart failure with preserved ejection fraction. Recent evidence includes myocardial infiltration, inducible nitric oxide synthase expression, patient phenogroups, and direct connections between comorbidities, inflammatory biomarkers, and abnormal myocardial structure/function.
