Glioblastoma and the search for non-hypothesis driven combination therapeutics in academia

Summary: Glioblastoma (GBM) is a cancer with high unmet clinical need. Traditional drug discovery methods have failed to overcome the challenges presented by tumor heterogeneity, therapeutic resistance, and tumor niches. Academic researchers are now implementing high throughput screening programs to identify novel combination targets for GBM by using more relevant phenotypic assays and focused libraries of small compounds. Recommendations are made to improve the preclinical small compound screening paradigm for better identification of tractable combinatorial inhibitors and drug targets specific to GBM.

Hallmarks of Cancer: New Dimensions

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

Target-specific compound selectivity for multi-target drug discovery and repurposing

Summary: Most drug molecules have the ability to modulate multiple target proteins, which can lead to both therapeutic effects and unwanted side effects. Evaluating the selectivity of a compound is an important factor in drug development and repurposing efforts. Traditional methods for characterizing selectivity fall short in quantifying how selective a compound is against a particular target protein. In this study, we propose an optimization-based selectivity scoring method that allows for the identification of potent and selective compounds against given kinase targets. We demonstrate the effectiveness of this method in finding highly selective compounds in computational experiments using a large-scale kinase inhibitor dataset.
