Improving ethanol tolerance of Saccharomyces cerevisiae through adaptive laboratory evolution using high ethanol concentrations as a selective pressure

Summary: This study describes an Adaptive Laboratory Evolution (ALE) strategy to develop Saccharomyces cerevisiae populations with increased tolerance to ethanol. The evolved populations after 100 generations could survive for 1 hour at 23% v/v ethanol and for 2 hours at 25% v/v ethanol. The fermentation ability and growth rate of the yeast strains were significantly improved through ethanol-based evolution.

The study investigated the use of consolidated bioprocessing (CBP) with a recombinant strain of S. cerevisiae AC14 capable of producing multiple lignocellulosic enzymes and fermenting xylose and glucose. The results were promising, with high ethanol productivity and complete hydrolysis of cellulose and xylan.

Genomic, transcriptomic, and metabolic characterization of 2-Phenylethanol-resistant Saccharomyces cerevisiae obtained by evolutionary engineering

Summary: This study aimed to obtain a 2-phenylethanol-resistant Saccharomyces cerevisiae strain by in vivo evolutionary engineering and characterize the adapted yeast at the genomic, transcriptomic and metabolic levels. Through gradual increase of 2-phenylethanol concentration in batch cultivations, an adapted strain that could tolerate 3.4 g/L of 2-phenylethanol was obtained. Genome sequencing and transcriptomic analysis revealed genetic mutations and upregulated genes associated with the adaptation process. These findings shed light on the resistance mechanism of the adapted yeast strain to 2-phenylethanol.

Saccharomyces cerevisiae surface display of endolysin LysKB317 for control of bacterial contamination in corn ethanol fermentations

Summary: Control of bacterial contamination in bioethanol fermentation facilities has traditionally relied on chemical-based products such as hop acids and use of antibiotics. Recently, a recombinant peptidoglycan hydrolase called LysKB317 was developed to inhibit Limosilactobacillus fermentum strains in corn mash fermentation. By anchoring LysKB317 on Saccharomyces cerevisiae EBY100 using cell surface display, it was demonstrated that the yeast surface-expressed endolysin significantly decreased bacterial cell counts and improved fermentation efficiency, providing potential alternative strategies for controlling microbial contamination.

Evolutionary and reverse engineering to increase Saccharomyces cerevisiae tolerance to acetic acid, acidic pH, and high temperature

Summary: This study used adaptive laboratory evolution (ALE) and whole-genome analysis to generate strains of Saccharomyces cerevisiae tolerant to high temperatures, acetic acid, and acidic pH. Through a year of evolution, two strains were selected, TAT12 and AT22, which were capable of growing and producing ethanol in conditions previously inhibitory to their ancestors. Whole-genome sequencing and transcriptomic analysis showed mutations in key genes related to stress response and replication, confirming the importance of these mutations in enhancing the tolerance of S. cerevisiae to harsh environments.
