Differential Recognition of Clinically Relevant Sporothrix Species by Human Mononuclear Cells

Summary: This study analyzed the cell wall composition of different morphotypes (germlings, conidia, and yeast-like cells) of Sporothrix globosa and compared the differences in cytokine production when these morphotypes interact with human peripheral blood mononuclear cells. The results showed that the cell wall composition and structure of the three Sporothrix species are different, affecting their interaction with human cells and generating species-specific cytokine profiles.

Summary: The cell wall of fungi is complex and consists of various crystalline and amorphous polysaccharides. Many enzymes are involved in the synthesis and degradation of these polysaccharides, and they play important roles in cell structure and function. The cell wall is a potential target for antifungal drugs, as its constituents are different from those in humans. Chitin, beta-glucans, and cellulose are the most common polysaccharides in the fungal cell wall. The hexosamine biosynthesis pathway is critical for cell wall formation, and GlcN-6-P synthase is an enzyme that has been considered as a potential target for antifungals.

Biofilm formation on cat claws by Sporothrix species: An ex vivo model

Summary: This study evaluated the ability of Sporothrix species to form biofilms on the surface of cat claws using an ex vivo model. Results showed that all species were able to form biofilms on cat claws, with metabolic activity similar to in vitro biofilms. This is the first report of an ex vivo biofilm model involving cat claws, which could increase the viable period of the fungus and the number of potentially infected animals and people.

Responses of Sporothrix globosa to the cell wall perturbing agents Congo Red and Calcofluor White

Summary: The study found that the dyes Congo Red (CR) and Calcofluor White (CFW) had an impact on the growth, morphology, and enzyme activity of Sporeothrix globosa fungus. Additionally, they induced yeast to transiently transform into conidia-forming filamentous cells, likely as a defense mechanism against the harmful effects of the dyes. The results shed light on the fungal responses to cell wall damage and proposed a hypothetical model based on membrane-spanning proteins known as mechanosensors.
