Fungal Whole-Genome Sequencing for Species Identification: From Test Development to Clinical Utilization

Population genomic analyses reveal evidence for limited recombination in the Candida auris in nature

Summary: Candida auris is a multidrug-resistant pathogenic yeast that has recently emerged as a global health threat. In this study, genetic analyses based on whole-genome sequences were conducted to investigate potential recombination events in natural populations of C. auris. The results revealed limited evidence for recombination both before and after the divergence of different clades within the species. However, the frequency of recombination was higher before the clade divergence and was observed to be limited in regions associated with antifungal resistance. These findings imply the possibility of ongoing genetic exchange in natural populations of this important yeast pathogen.

Factors that influence bidirectional long-tract homozygosis due to double-strand break repair in Candida albicans

Summary: Genomic rearrangements in Candida albicans are often the by-product of DNA break repair, such as repair through homologous recombination resulting in loss-of-heterozygosity (LOH). Mechanisms like break-induced replication (BIR) and mitotic crossover (MCO) can lead to long LOH tracts ascending towards the centromere. The study of DNA repair mutants in C. albicans shows differences in genome stability compared to model organism Saccharomyces cerevisiae, and suggests that LOH can affect alleles near the centromere from DNA break sites.
