Evolving concepts in the pathophysiology of atherosclerosis: from endothelial dysfunction to thrombus formation through multiple shades of inflammation

Giovanni Cimmino et al.

Summary: Atherosclerosis is the underlying cause of most cardiovascular diseases, and the role of immunity and inflammation in its formation and complications is well established. In addition to conventional risk factors, nonconventional factors such as gut microbiota may also play a role in disease progression, and targeting inflammation appears to be a promising approach for managing atherosclerosis-related diseases.

Insights into the development of insulin resistance: Unraveling the interaction of physical inactivity, lipid metabolism and mitochondrial biology

Rachel M. Handy et al.

Summary: Impairments in peripheral tissue insulin signaling, caused by a high-lipid environment and increased mitochondrial reactive oxygen species production, are believed to contribute to insulin resistance and type 2 diabetes. However, physical inactivity can also promote insulin resistance through alternative mechanisms, such as a reduction in protein synthesis. Mitochondrial biology plays a crucial role in the interaction between physical (in)activity, lipid metabolism, and insulin signaling.

Summary: Insulin resistance (IR) is crucial in metabolism-related diseases and provides a basis for understanding chronic diseases. This study reviewed the causes, mechanisms, and treatments of IR. Various factors including genetics, obesity, and disease lead to abnormalities in the insulin signaling pathway and development of IR. Therapeutic strategies include exercise, dietary changes, chemotherapy, and traditional Chinese medicine treatments. Further research is needed to define biomarkers and explore targeted drug treatments for IR to improve patients' quality of life.

Celestino Sardu et al.

Summary: This study evaluated the expression of SGLT2, inflammatory cytokines, and sirtuins in breast fat tissue and serum cytokines of pre-menopausal women. The results showed that women with fatty breasts over-expressed SGLT2 and inflammatory cytokines, while downregulating breast sirtuins. The study also found correlations between the expression of SGLT2/cytokines/sirtuins and clinical variables related to cardiovascular health.

Uric acid plasma levels are associated with C-reactive protein concentrations and the extent of coronary artery lesions in patients with acute coronary syndromes

Giovanni Cimmino et al.

Summary: Many studies have found that inflammation is a key player in the development of acute coronary syndromes (ACS) as it impairs endothelial function and promotes atherothrombotic events. Recent evidence suggests that elevated levels of uric acid (UA) in the blood may be a risk factor for ACS. This study investigated the relationship between UA levels, inflammation, and the extent of coronary atherosclerosis in ACS patients. The results showed that ACS patients with higher UA levels had elevated levels of C-reactive protein and presented with more severe coronary disease.

Gut Dysbiosis and Immune System in Atherosclerotic Cardiovascular Disease (ACVD)

Ji Youn Yoo et al.

Summary: Atherosclerosis, a major cause of cardiovascular disease, can be influenced by alterations in the gut microbiota composition, leading to gut dysbiosis. This dysbiosis activates immune processes and systemic inflammation, contributing to the development of atherosclerotic cardiovascular disease (ACVD). The gut microbiota produces metabolites, such as trimethylamine (TMA) and short-chain fatty acids (SCFAs), that can either promote or decrease atherogenesis. Understanding the relationship between the gut microbiota and the host immune system could provide insights into ACVD development, prognostic factors, and potential treatments.

Summary: The relationship between sleep duration and cardiovascular diseases has been explored in observational studies, but the causality remains unclear. This meta-analysis examined the impact of sleep duration on the risk of cardiovascular diseases using observational and Mendelian randomization studies. The results suggest that short sleep duration may be a causal risk factor for coronary artery disease and heart failure, while long sleep duration is associated with the risk of coronary artery disease, stroke, and mortality, but lacks causal associations in Mendelian randomization studies. Increasing sleep duration may benefit cardiovascular health.
