Two-year clinical outcomes of triple therapy with photodynamic therapy, anti-vascular endothelial growth factor agent, and triamcinolone acetonide for neovascular age-related macular degeneration

Ryo Ando et al.

Summary: This study retrospectively analyzed the 2-year treatment outcomes of triple therapy for nAMD in Japanese patients. The results showed that triple therapy could effectively improve BCVA and CRT, with approximately 40% of patients not requiring retreatment within 2 years. The type of anti-VEGF agents used did not significantly affect the treatment outcomes.

Towards a better understanding of non-exudative choroidal and macular neovascularization

Riccardo Sacconi et al.

Summary: Non-exudative macular and choroidal neovascularization (MNV and CNV) refers to treatment-naive type 1 neovascularization without exudation. This entity was first reported in the late 90s and has been increasingly recognized with advanced retinal imaging. However, there is a lack of consensus regarding the definition and clinical management. This review discusses the prevalence, imaging features, clinical course, and response to treatment of non-exudative MNV and CNV, providing key insights and proposing a new theory about their origin and course.

Lycopene inhibits endothelial-to-mesenchymal transition of choroidal vascular endothelial cells in laser-induced mouse choroidal neovascularization

Lele Li et al.

Summary: Choroidal neovascularization (CNV) is a leading cause of blindness, and LYC has been found to inhibit the process of EndMT in choroidal vascular endothelial cells (CVECs). This inhibition occurs through the modulation of the AR/MITF/PEDF/LR/AKT/beta-catenin pathway. Additionally, LYC has shown promising therapeutic effects in a mouse model of CNV.

Summary: Neovascular age-related macular degeneration (nAMD) is a leading cause of visual impairment and blindness, but current anti-VEGF therapies have limitations. This article discusses the need for new and improved therapies for nAMD and explores the efficacy of multi-targeted therapies that target both the VEGF pathway and other pathways.

Highly Retina-Permeating and Long-Acting Resveratrol/Metformin Nanotherapeutics for Enhanced Treatment of Macular Degeneration

Duc Dung Nguyen et al.

Summary: We developed a nanomedicine strategy to create nanotherapeutics with high retinal permeability and sustained bioactive delivery. The nanotherapeutics were designed through aminolysis of resveratrol-encapsulated polycaprolactone nanoparticles, followed by the formation of amide linkages with a transacting activator of transcription cell penetrating peptide and metformin. In vitro and in vivo studies demonstrated the effectiveness of the nanotherapeutics in improving retinal permeability and targeting retinal diseases.

Animal Models of Choroidal Neovascularization: A Systematic Review

Bjorn K. Fabian-Jessing et al.

Summary: This systematic review provides a detailed overview of contemporary animal models of choroidal neovascularization (CNV). The study found that mice are the most commonly used animal model, with laser-induced mouse model being the predominant model for experimental CNV. However, the widespread use of young, healthy male animals may complicate clinical translation, and inadequate reporting challenges reproducibility.

Shikonin alleviates choroidal neovascularization by inhibiting proangiogenic factor production from infiltrating macrophages

Ying Wang et al.

Summary: Choroidal neovascularization (CNV) is a major cause of vision loss in elderly individuals with age-related macular degeneration (AMD). Shikonin (SHI), a natural compound extracted from Chinese herb radix arnebiae, has been found to inhibit CNV lesion development and macrophage infiltration by targeting the PKM2/ STAT3/CD163 pathway. This study suggests SHI as a potential novel therapy for CNV by suppressing pro-angiogenic cytokine secretion from macrophages.

Transcriptomics analysis of Ccl2/Cx3cr1/Crb1rd8 deficient mice provides new insights into the pathophysiology of progressive retinal degeneration

Anna Badia et al.

Summary: Chronic oxidative stress and immune dysregulation play crucial roles in the pathogenesis of most retinal degenerative diseases, including age-related macular degeneration. The DKOrd8 mouse model demonstrates early changes in the retina, such as photoreceptor atrophy and pigment alterations, leading to impaired retinal function. Transcriptomic analysis revealed significant differential expression of genes related to oxidative stress and neuronal function, emphasizing the relevance of these processes in pathological retinal degeneration.
