p75NTR enhances cognitive dysfunction in a mouse Alzheimer's disease model by inhibiting microRNA-210-3p-mediated PCYT2 through activation of NF-ΚB

Zhongliang Wei et al.

Summary: This study investigated the role of p75 neurotrophin receptor (p75NTR) in cognitive dysfunction in Alzheimer's disease (AD). In vivo and in vitro models of AD were established using C57BL/6 mice and neuroblastoma cells treated with amyloid-beta1-42 (Aβ1-42), respectively. RNA-sequencing and bioinformatics analysis predicted the downstream genes of p75NTR. The interaction between p75NTR, nuclear factor kappa B (NF-κB), microRNA-210-3p (miR-210-3p), and phosphoethanolamine cytidylyltransferase 2 (PYCT2) was verified, and their effects on cognitive behaviors and biological characteristics of hippocampal neurons in AD mouse models were analyzed. p75NTR knockout alleviated cognitive dysfunction and reduced hippocampal neuron damage and apoptosis in AD-like mice. p75NTR upregulated miR-210-3p expression by activating NF-κB, thereby limiting PCYT2 expression. PCYT2 silencing in p75NTR-/- mice promoted neuronal apoptosis and aggravated cognitive dysfunction in AD mouse models. In summary, p75NTR accelerates cognitive dysfunction in AD through the NF-κB/miR-210-3p/PCYT2 axis.

Identification of altered exosomal microRNAs and mRNAs in Alzheimer's disease

Lining Su et al.

Summary: The study examined the expression patterns of mRNAs and miRNAs in exosomes derived from Alzheimer's disease (AD) and healthy mice, finding differential expression of 1320 mRNAs and 29 miRNAs between the two groups. Key genes such as Chi3l1 and Rhog were validated to be dysregulated in AD mice, along with miR-148a-5p and miR-27a-5p. GO and KEGG analysis helped determine the functions of the differentially expressed mRNAs and potential target genes of miRNAs. Through the ceRNA hypothesis, an integrated ceRNA network of circRNA-lncRNA-miRNA-mRNA was established, indicating the involvement of exosomal lncRNAs, mRNAs, circRNAs, and miRNAs in the progression of AD, potentially serving as biomarkers and therapeutic targets.

The p38MAPK-MK2 Signaling Axis as a Critical Link Between Inflammation and Synaptic Transmission

Edward Beamer et al.

Summary: MK2, a downstream target of p38(MAPK), regulates gene expression and synaptic plasticity indirectly through control of RNA-binding proteins and cytoskeletal dynamics. While its role in inflammation and cellular stress is well-described, the involvement of neuronal MK2 in mediating synaptic plasticity in response to inflammation remains unclear.

Therapeutic Potential of Vital Transcription Factors in Alzheimer's and Parkinson's Disease With Particular Emphasis on Transcription Factor EB Mediated Autophagy

Sachchida Nand Rai et al.

Summary: Autophagy is essential for cellular homeostasis, and dysregulation in this pathway is implicated in neurodegenerative disorders. TFEB, a key regulator of autophagy and lysosomal biogenesis, is considered a potential therapeutic target for treating these disorders. Studies have shown that targeting TFEB may help alleviate neurotoxicity and rescue neurodegeneration in animal models, providing insights for potential therapeutic strategies in neurodegenerative disorders.

Electroacupuncture Improves M2 Microglia Polarization and Glia Anti-inflammation of Hippocampus in Alzheimer's Disease

Lushuang Xie et al.

Summary: Electroacupuncture treatment improves cognition and memory in AD rats, inhibits glial cell activation, promotes microglial polarization towards the M2 phenotype, reduces pro-inflammatory cytokine expression, and increases anti-inflammatory cytokine expression. Additionally, electroacupuncture affects immune responses by inhibiting the NF-kappa B pathway and activating the Stat6 pathway.

MiR-191-5p alleviates microglial cell injury by targeting Map3k12 (mitogen-activated protein kinase kinase kinase 12) to inhibit the MAPK (mitogen-activated protein kinase) signaling pathway in Alzheimer's disease

Wenjun Wan et al.

Summary: The study revealed that miR-191-5p was downregulated in A beta 1-42-stimulated microglia and hippocampal tissues of APP/PS1 mice, and overexpression of miR-191-5p alleviated A beta 1-42-induced microglia cell injury.

Rui Duan et al.

Summary: The Ang-(1-7) analogue AVE0991 inhibits astrocyte-mediated neuroinflammation via the SNHG14/miR-223-3p/NLRP3 pathway and provides neuroprotection in APP/PS1 mice. These findings reveal the mechanisms by which Ang-(1-7) inhibits neuroinflammation in Alzheimer's disease and highlight the potential of AVE0991 as a treatment for AD.

CircPTK2-miR-181c-5p-HMGB1: a new regulatory pathway for microglia activation and hippocampal neuronal apoptosis induced by sepsis

Min Li et al.

Summary: The study demonstrates the regulatory relationship between circPTK2, miR-181c-5p, and HMGB1 in microglia and sepsis mice. HMGB1 and circPTK2 were upregulated, while miR-181c-5p was downregulated in response to LPS induction in microglia. Targeting circPTK2/miR-181c-5p/HMGB1 signaling pathway may offer potential therapeutic targets for sepsis-induced cognitive dysfunction and neuronal apoptosis.

Selective removal of astrocytic APOE4 strongly protects against tau-mediated neurodegeneration and decreases synaptic phagocytosis by microglia

Chao Wang et al.

Summary: Removing astrocytic APOE4 significantly reduces tau-mediated neurodegeneration, decreases tau pathology, lowers the levels of phosphorylated tau, and diminishes disease-associated gene expression signatures in neurons, oligodendrocytes, astrocytes, and microglia. This suggests a crucial role for astrocytic apoE in synaptic degeneration and points to the potential therapeutic implications of targeting this pathway in Alzheimer's disease.
