Purification, full-length sequencing and genomic origin mapping of eccDNA

Yuangao Wang et al.

Summary: A new three-step eccDNA purification (3SEP) procedure is proposed to efficiently purify eccDNA with high purity. In addition, a full-length eccDNA sequencing technique is developed, along with an eccDNA caller software, for accurate identification and mapping of eccDNA. These methods will facilitate the study of eccDNA and have potential diagnostic and clinical applications.

Long non-coding RNA CCL14-AS suppresses invasiveness and lymph node metastasis of colorectal cancer cells by regulating MEP1A

Mingzhou Li et al.

Summary: By analyzing the TCGA dataset, we identified a novel lncRNA called CCL14-AS, which is enriched in the cytoplasm and negatively correlated with lymph node metastasis and unfavorable prognosis of colorectal cancer (CRC). Functional experiments and mouse models confirmed the inhibitory role of CCL14-AS in CRC. Our findings suggest that the CCL14-AS/MEP1A axis serves as a critical regulator in CRC progression and may be a potential biomarker and therapeutic target in advanced CRC.

Identification and Characterization of Extrachromosomal Circular DNA in Plasma of Lung Adenocarcinoma Patients

Xiaoqiong Wu et al.

Summary: The study identified cancer-specific eccDNA in the plasma of lung adenocarcinoma patients, potentially serving as promising biomarkers for early diagnosis. A comparison with healthy individuals and healthy gravidas revealed four eccDNA uniquely expressed in lung adenocarcinoma patients, which could be potential markers for early detection.

Characteristics of Fetal Extrachromosomal Circular DNA in Maternal Plasma: Methylation Status and Clearance

Sarah T. K. Sin et al.

Summary: The research found that fetal eccDNA in plasma is relatively hypomethylated compared to maternal eccDNA. The methylation densities of eccDNA are positively correlated with their sizes. Fetal eccDNA is rapidly cleared from maternal blood after delivery, similar to fetal linear DNA.

MicroDNA levels are dependent on MMEJ, repressed by c-NHEJ pathway, and stimulated by DNA damage

Teressa Paulsen et al.

Summary: This study suggests that microDNA levels are influenced by resection after double-strand DNA break (DSB) and repair by Microhomology Mediated End Joining (MMEJ). The accumulation of microDNA occurs during DNA repair pathways following endogenous or induced DNA breaks.

Small extrachromosomal circular DNA (eccDNA): major functions in evolution and cancer

Xiaoxuan Ling et al.

Summary: eccDNA is a type of circular DNA that is independent of chromosomes but plays important roles in monitoring, early diagnosis, treatment, and prediction of cancer. They are commonly found in both normal and cancer cells, functioning through various mechanisms in stress response, aging, carcinogenesis, and drug resistance during cancer treatment.
