Liver ductal organoids reconstruct intrahepatic biliary trees in decellularized liver grafts

Summary: In this study, researchers reconstructed intrahepatic bile ducts in a rat decellularized liver graft using organoid technology. Through an ex vivo perfusion culture system, they successfully demonstrated the biliary characteristics of repopulated liver organoids and reconstructed biliary tree-like networks with luminal structures. They also established a method to mimic the appropriate cell distribution in the native liver and demonstrated reconstructed bile ducts in human organoids.

Human branching cholangiocyte organoids functional bile duct formation

These organoids show high similarity to primary cholangiocytes, and the branching growth process is dependent on JAG1/NOTCH2 signaling. When applied to cholangiocarcinoma tumor organoids, the branching cholangiocarcinoma organoids exhibit morphology and transcriptomic profile closer to primary tumors and increased chemoresistance.

This study demonstrates a biocompatible stereolithographic approach to encapsulate cholangiocytes in hydrogel structures, guiding them to form branched tubular networks. The resulting structures, resembling those found in the biliary tree, show stable organization and epithelial maturation markers without the need for additional supporting materials.
