Progress in the studies on the molecular mechanisms associated with multidrug resistance in cancers

Lei Zhang et al.

Summary: Chemotherapy is an important method for treating cancer, but multi-drug resistance (MDR) is a major reason for its failure. Almost all anti-tumor drugs develop drug resistance over time, reducing their effectiveness in killing cancer cells. Chemoresistance can lead to rapid cancer recurrence and patient death.

Advanced techniques for gene heterogeneity research: Single-cell sequencing and on-chip gene analysis systems

Zaizai Dong et al.

Summary: Gene heterogeneity has a significant impact on cellular behaviors and poses challenges to biomedicine. Traditional gene analysis methods obscure the gene properties of individual cells, while single-cell gene analysis techniques offer unprecedented opportunities for high precision and high throughput research on gene heterogeneity. This review summarizes the latest techniques in single-cell sequencing and on-chip gene analysis, highlighting their principles and application scenarios in gene heterogeneity research. The future challenges and directions for technical improvement and clinical applications of single-cell gene analysis techniques are also discussed.

Decoding dynamic epigenetic landscapes in human oocytes using single-cell multi-omics sequencing

Rui Yan et al.

Summary: The research developed a novel single-cell multi-omics sequencing method and found that DNA methylation in human oocytes increases globally during growth, correlating with chromatin accessibility, while specific features of DNA methylation increase with active transcription. Integrated analysis of multi-omics data revealed species-specific gene expression and promoter accessibility contributing to gene body methylation programs.

SMOOTH-seq: single-cell genome sequencing of human cells on a third-generation sequencing platform

Xiaoying Fan et al.

Summary: There is currently no effective method to detect structure variations (SVs) and extra-chromosomal circular DNAs (ecDNAs) at single-cell whole-genome level. However, the newly developed SMOOTH-seq method shows promise in reliably and effectively detecting SVs and ecDNAs in individual cells, while having relatively limited accuracy in detecting CNVs and SNVs. SMOOTH-seq opens up new possibilities for single-cell whole-genome sequencing by providing high fidelity reads of kilobases long.
