Charting co-mutation patterns associated with actionable drivers in intrahepatic cholangiocarcinoma

Futibatinib for FGFR2-Rearranged Intrahepatic Cholangiocarcinoma

Targeting BRCA-mutant biliary tract cancer: Current evidence and future perspectives

Summary: Biliary tract cancers (BTCs) are aggressive tumors of the biliary system that are often diagnosed at an advanced stage with a poor prognosis. BRCA1/2 mutations have been reported in BTC patients, and treatment with PARP inhibitors has shown some effectiveness without severe side effects. The frequencies and types of BRCA1/2 mutations in advanced BTCs vary among studies. Differences in the therapeutic effects and response to immunotherapy between germline and somatic BRCA mutations have been observed.

Rapid and Cost-Efficient Detection of RET Rearrangements in a Large Consecutive Series of Lung Carcinomas

Summary: RET-kinase-activating gene rearrangements are found in 1-2% of non-small-cell lung carcinomas (NSCLCs), with reliable detection requiring next-generation sequencing (NGS). A new assay based on the comparison of RNA transcripts of different portions of the RET gene was developed, allowing the detection of RET translocations in NSCLCs. The study found a significant occurrence of RET rearrangements in EGFR/KRAS/ALK/ROS1/BRAF/MET-negative carcinomas and a correlation between RET rearrangement and thymidylate synthase expression in female patients.

Durable Response after Olaparib Treatment for Perihilar Cholangiocarcinoma with Germline BRCA2 Mutation

Recent studies have identified actionable genomic alterations, such as homologous recombination deficiency (HRD), that can affect treatment response. This case report discusses a patient with BRCA2-mutant CCA who showed a durable response to olaparib, a PARP inhibitor, after intolerable toxicity with gemcitabine/cisplatin. Further clinical trials are needed to confirm the role of PARP inhibition in similar patients.
