Effects of Dimerization, Dendrimerization, and Chirality in p-BthTX-I Peptide Analogs on the Antibacterial Activity and Enzymatic Inhibition of the SARS-CoV-2 PLpro Protein

Development of Antiplasmodial Peptide-Drug Conjugates Using a Human Protein-Derived Cell-Penetrating Peptide with Selectivity for Infected Cells

Summary: Targeted drug delivery with peptide-drug conjugates (PDCs) was investigated as an alternative antimalarial therapy. PDCs with low micromolar potency were developed by conjugating a synthetic peptide derived from an innate human defense molecule with the antimalarial drug primaquine (PQ). Various PDCs were designed to identify the optimal conjugation site and investigate linker length, hydrophilicity, and cleavability. Conjugation within a flexible spacer region of the peptide, with a cleavable linker to liberate the PQ cargo, was crucial for retaining the activity of both the peptide and drug.

Antiviral Evaluation of New Synthetic Bioconjugates Based on GA-Hecate: A New Class of Antivirals Targeting Different Steps of Zika Virus Replication

Summary: Re-emerging arboviruses, particularly the Zika virus outbreak in South and Central America, pose a serious health threat in urban tropical areas. In this study, a peptide derived from human blood serum, Gallic acid-Hecate, was shown to effectively inhibit the replication of Zika virus in cultured cells. One of the synthesized metabolites of Gallic acid-Hecate, GA-metabolite 5, demonstrated the highest inhibitory activity against two strains of Zika virus. Furthermore, GA-metabolite 5 did not affect cell growth, making it a promising candidate for the development of antiviral drugs.

Conjugates for use in peptide therapeutics: A systematic review and meta-analysis

Advances and perspectives for antimicrobial peptide and combinatory therapies

The membrane-specificity of AMPs and their potential to combat drug-resistant microbes provide hope in the medical and therapeutic field. The high global death rate due to antimicrobial resistance (AMR) has led to the search for new molecules and product developments, primarily antibiotics. In addition to using multiple drugs, the synergistic action of AMPs and current antibiotic therapy has shown promise in treating difficult-to-treat infectious diseases caused by bacteria. This review highlights the recent advancements in medical and antibiotic therapy using AMP-based products and discusses the potential effects of using AMPs in combination with current drugs for the treatment of bacterial infectious diseases.
