Summary: Two novel series of benzimidazole-triazole hybrids were synthesized and evaluated as multi-target inhibitors against EGFR, VEGFR-2, and Topo II. Compounds 5a and 6g showed the highest anticancer activity against four cancer cell lines and were further studied for their inhibition of EGFR, VEGFR-2, and Topo II. Compound 5a exhibited good EGFR inhibitory activity, moderate VEGFR-2 inhibitory activity, and strong Topo II inhibitory activity.

Summary: This study found that thymol (THY) from thyme species can inhibit oxidative stress, inflammation, and lung fibrosis-related miR-29a/TGF-beta and PI3K/phospho-Akt signaling pathways. The results showed that THY effectively alleviated pulmonary fibrosis, upregulated miR-29a expression, and downregulated TGF-beta and PI3K/Akt signaling pathways. These findings are worthy of further investigation as a possible mechanism for THY's anti-fibrotic effects.

An emerging flavone glycoside from Phyllanthus emblica L.: As promiscuous enzyme inhibitor and potential therapeutic in chronic diseases

Summary: In current drug discovery, there is a growing focus on designing drug molecules that interact with multiple targets simultaneously (exhibiting compound promiscuity). Previous screening research on 54 medicinal plants found that the methanol extract of Phylanthus emblica L. leaves showed promising inhibitory activity against alpha amylase, tyrosinase, and hyaluronidase enzymes. Further fractionation of the extract led to the isolation of two flavone glycosides, with one of them, luteolin-7-O-a-L-rhamnoside, exhibiting strong inhibitory activity against the enzymes. This study suggests that luteolin-7-O-a-L-rhamnoside could be used as a potential promiscuous enzyme inhibitor for the management and treatment of certain chronic diseases.

Triazole-tethered boswellic acid derivatives against breast cancer: Synthesis, in vitro, and in-silico studies

Summary: A series of new analogues of 3-O-acetyl-11-keto-fi-boswellic acid and 3-O-acetyl-fi-boswellic acid were synthesized using Click chemistry reaction protocol. The synthesized compounds showed potent anti-proliferative activity against triple-negative breast cancer cell lines. Compounds 10j, 10f, and 10i exhibited exceptional inhibitory potency and were more potent than the parent compounds. The cheminformatics method identified CHK1 as the potential drug target for the most potent compounds.

Confusoside from Anneslea fragrans Alleviates Acetaminophen-Induced Liver Injury in HepG2 via PI3K-CASP3 Signaling Pathway

Summary: The study demonstrates that Confusoside (CF), a major chemical compound in Anneslea fragrans Wall. leaves, has remarkable antioxidant and anti-inflammatory effects, and also exhibits hepatoprotective effect against acetaminophen (APAP)-induced hepatic injury. CF reduces inflammation and inhibits cell apoptosis, significantly decreasing the levels of liver injury biomarkers and inflammatory cytokines, inhibiting the activation of the NF-kappa B signaling pathway, and regulating the expression of apoptosis pathway-related proteins. These findings suggest that CF could be a potential reagent for the prevention and treatment of APAP-induced liver injury.

Microbial Transformation of the Sesquiterpene Lactone, Vulgarin, by Aspergillus niger

Summary: The biotransformation of vulgarin, a sesquiterpene lactone obtained from Artemisia judaica, was performed by Aspergillus niger to produce three more polar metabolites. The structures of these metabolites were determined using NMR spectroscopy and HRESIMS. The anti-inflammatory activity of vulgarin and its metabolites was evaluated, and vulgarin showed preferential inhibitory activity against COX-2, while its metabolite (3a) exhibited selective inhibition against COX-1.

Bio-evaluation of Untapped Alkaloids from Vinca minor Enriched by Methyl-jasmonate-induced Stress: an Integrated Approach

Summary: The modulation of biosynthetic pathways by stimulating stress-induced responses in plants has been proven to be an effective way to diversify known natural products. In this study, the effect of methyl jasmonate (MeJA) on Vinca minor alkaloids distribution was investigated, and three compounds were successfully isolated and subjected to bioassays. The extracts and compounds showed antimicrobial and cytotoxic activities, as well as potential wound healing effects through the TGF-beta pathway. The expression of markers related to this pathway and wound healing was assessed, and molecular docking provided insights into the binding ability of the compounds with different active sites in mTOR.

Enzymatic Synthesis of Thymol Octanoate, a Promising Hybrid Molecule

Summary: Interest in the synthesis and application of thymol esters has increased recently due to its various biological activities. This study explored the enzymatic synthesis of thymol octanoate using soluble lipases and immobilized lipase biocatalysts. Candida antarctica lipase B in its soluble form was found to be the most active biocatalyst. Different feeding strategies were evaluated, and the optimal reaction condition was determined to be a thymol/acid molar ratio of 1:4 mol/mol. This study demonstrated the potential of using the enzyme in successive reaction cycles.

It modulated oxidative stress, neurotransmitters, and brain ATP levels, and ameliorated histopathological damage, astrocyte swelling, and brain edema. Thymol also downregulated NF-kB and upregulated GFAP expression, and promoted CREB and BDNF expression, as well as increased brain cAMP levels.
