Effect of Phosphorylation Sites Mutations on the Subcellular Localization and Activity of AGPase Bt2 Subunit: Implications for Improved Starch Biosynthesis in Maize

Preparation of Polyclonal Antibody against ZmBT1 Protein and Its Application in Hormone-Regulated Starch Synthesis

Summary: To investigate the role of ZmBT1 in starch accumulation during maize grain development and its expression and distribution in different maize tissues, a polyclonal antibody was prepared. The recombinant plasmid pGEX-6p-ZmBT1-C (382-437aa) was successfully expressed and Gst-ZmBT1-C purified as the antigen for antibody preparation. The results confirmed specific recognition of ZmBT1 protein in maize tissues by the ZmBT1 antibody. Western blotting and quantitative real-time PCR showed that ZmBT1 expression varied among tissues, with the highest content in the grain and endosperm. Furthermore, ZmBT1 expression was influenced by plant hormones, indicating its critical role in starch accumulation.

Comparative Study of Starch Phosphorylase Genes and Encoded Proteins in Various Monocots and Dicots with Emphasis on Maize

Summary: This study reports the structure, function, and evolution of genes encoding PHO1 and PHO2, and their ligand-binding sites in monocots and dicots. The genes controlling PHO2 were found to be more conserved than those controlling PHO1, and cis-regulatory elements in the promoter region of both genes were identified. Results showed that ABA could up-regulate the expression of both PHO1 and PHO2 in Zea mays.
