Plant protoplasts in the age of synthetic biology

Summary: This review discusses the role of protoplasts in the 'design-build-test-learn' cycle in synthetic biology and their potential impact on other areas of research. Protoplasts, plant cells with their cell walls removed, have been used for genetic transformation and studying plant physiology and genetics for decades. With the emergence of synthetic biology, protoplasts play a crucial role in speeding up the research cycle. However, challenges remain in fully exploiting their potential in synthetic biology.

Combining multiplex gene editing and doubled haploid technology in maize

Summary: A rapid and efficient strategy was developed to produce genetically identical plants carrying various combinations of homozygous edits for functional analysis. This strategy combines highly multiplex gene editing in maize with in vivo haploid induction and efficient in vitro generation of doubled haploid plants. Multiple homozygous lines with different combinations of edits were obtained within three generations, showing reproducible phenotypical differences. This approach is expected to facilitate the study of gene families and improve quantitative crop traits.

Summary: In this study, different delivery methods of CRISPR/Cas9 reagents were compared for their efficiency in genome editing of chicory, as well as the frequency of off-target mutations and their environmental and economic impacts. The study found that using RNPs as the delivery method resulted in high editing efficiency, no off-target mutations, and avoided the integration of unwanted plasmid DNA and the need for gene segregation.

Genetic Manipulation and Bioreactor Culture of Plants as a Tool for Industry and Its Applications

Research has resulted in efficient plant systems that can produce specific metabolites and proteins, and the use of bioreactors enables industrial-scale production. This review paper provides an overview of the key research in the past decade, highlighting the use of transgenic plants as small, green biofactories and discussing the current state of patents in this field.

Anatomical and hormonal factors determining the development of haploid and zygotic embryos of oat (Avena sativa L.)
