DeepTP: A Deep Learning Model for Thermophilic Protein Prediction

Summary: Thermophilic proteins are valuable in biopharmaceuticals and enzyme engineering. To address the limitations of existing prediction models, a deep learning model called DeepTP was proposed, which utilized self-attention and multiple-channel feature fusion. DeepTP outperformed existing methods in performance comparison, achieving AUC values of 0.944 and 0.801 in independent balanced test set and validation set, respectively. In the unbalanced test set, DeepTP had an average precision (AP) of 0.536. The tool is freely available.

Characterization of high-yield Bacillus subtilis cysteine protease for diverse industrial applications

Summary: Bacterial proteases were isolated from skimmed milk agar media through serial dilution. The strains were identified as Bacillus subtilis and designated as A1 (MT903972), A2 (MT903996), A4 (MT904091), and A5 (MT904796) based on microbial biomass production, biochemical tests, protease-specific activity, and 16s RNA gene sequencing. Among them, A4 Bacillus subtilis exhibited the highest protease-specific activity of 76,153.84 U/mg. The identified cysteine protease showed great potential in various industrial applications.

Loop optimization of Trichoderma reesei endoglucanases for balancing the activity-stability trade-off through cross-strategy between machine learning and the B-factor analysis

Summary: In this study, a cross-strategy combining machine learning and B-factor analysis was used to predict and optimize the thermostability and activity of Trichoderma reesei endoglucanases. The results showed that the optimized enzymes not only improved enzymatic activity but also had better thermal properties compared to the wild type. The study suggests that loop optimization is crucial for balancing the activity-stability trade-off and provides new insights into the interaction between loop region function and enzymatic characteristics.
