Lipidomic analysis of brain and hippocampus from mice fed with high-fat diet and treated with fecal microbiota transplantation

Jinchen Li et al.

Summary: This study aimed to investigate whether dietary fatty acids affect brain lipids through gut microbiota. The results showed that high-fat diet and fecal microbiota transplant had certain effects on the content and composition of fatty acids in the brain, especially glycerol phospholipids. Acyl-carnitines (AcCa) and lysophosphatidylgylcerol (LPG) increased in all types of high-fat diet groups, while phosphatidic acids (PA), phosphatidylethanolamine (PE) and sphingomyelin (SM) significantly increased in the n-6 polyunsaturated fatty acid (n-6 PUFA) group. By altering the fecal microbiota, dietary fatty acids may affect brain lipids.

Effects of Different Storage Conditions on Lipid Stability in Mice Tissue Homogenates

Erika Dorochow et al.

Summary: Lipids are crucial biomolecules with involvement in various physiological and pathological processes, making the elucidation of their presence in tissue samples highly important. However, tissue analysis presents challenges, as pre-analytical factors can significantly alter lipid concentration, affecting the reliability of research results. This study investigates the influence of pre-analytical factors on lipid profiles in homogenized tissues. The results suggest that swift processing of tissue homogenates under cooled conditions is a viable option for reliable lipid analysis.

Benchmarking One-Phase Lipid Extractions for Plasma Lipidomics

Marcus Hoering et al.

Summary: This study evaluated the lipid recovery of popular one-phase extraction methods for human plasma samples and found that the recovery of polar lipid classes was sufficient, while the recovery of nonpolar lipid classes was low. Additionally, precipitation of lipids was observed in some solvents. Therefore, the solvent polarity and lipid class should be considered when performing lipid extractions.

Determination of tissue contributions to the circulating lipid pool in cold exposure via systematic assessment of lipid profiles

Raghav Jain et al.

Summary: Plasma lipid levels are complex and regulated by tissues such as liver, brown adipose tissue, kidney, and intestine. Using lipidomics, researchers found that acylcarnitines and ceramides in plasma are significantly altered by acute cold exposure, with liver and brown adipose tissue identified as major contributors. The study also identified kidney and intestine as novel contributors to the plasma lipid pool, highlighting their importance in understanding lipid functions in metabolic diseases.

Application of Lipid Class Ratios for Sample Stability Monitoring-Evaluation of Murine Tissue Homogenates and SDS as a Stabilizer

Sabrina Krautbauer et al.

Summary: In this study, the stability of lipid profiles in tissue homogenates was investigated, with ratios of lipid classes used to evaluate sample stability and the impact of stabilizers. The addition of SDS showed concentration-dependent stabilization of lipid profiles in some tissues, while causing an increase in a specific lipid ratio in others. These findings demonstrate the potential of lipid class ratios to assess sample stability and the effects of stabilizing agents.
