Roles of Hydrogen Sulfide in Hypertension Development and Its Complications: What, So What, Now What

Rui Wang

Summary: Hypertension is a complex health challenge with multiple causes, while hydrogen sulfide (H2S) serves as a multifunctional gasotransmitter. Animal studies conducted 15 years ago have established the critical role of endogenous H2S deficiency in hypertension development, leading to further investigations into its cardiovascular effects and underlying molecular and cellular mechanisms. We are also beginning to understand the involvement of altered H2S metabolism in human hypertension. This article aims to provide a comprehensive understanding of H2S's roles in hypertension development in both animals and humans, and review H2S-based antihypertension therapeutic strategies.

Sodium Thiosulfate Improves Hypertension in Rats with Adenine-Induced Chronic Kidney Disease

Chien-Ning Hsu et al.

Summary: This study investigated the effects of hydrogen sulfide and its derivative therapy on hypertension induced by chronic kidney disease. The results showed that oral administration of sodium thiosulfate could reduce blood pressure and increase plasma levels of hydrogen sulfide and thiosulfate. Additionally, thiosulfate therapy restored nitric oxide bioavailability and rebalanced the renin-angiotensin system.

Development of Hydropersulfide Donors to Study Their Chemical Biology

Vinayak S. Khodade et al.

Summary: Researchers have developed small-molecule donors to efficiently release Hydropersulfides (RSSH) and uncover their potential physiological functions. The complex relationship among RSSH, hydrogen sulfide (H2S), and higher order polysulfides in biological systems poses challenges in distinguishing their activities. Further investigation is needed to understand the biological responses and develop optimal RSSH donors for specific tissues/pathologies.

Dietary Supplementation with Cysteine during Pregnancy Rescues Maternal Chronic Kidney Disease-Induced Hypertension in Male Rat Offspring: The Impact of Hydrogen Sulfide and Microbiota-Derived Tryptophan Metabolites

Chien-Ning Hsu et al.

Summary: This study found that supplementing l- or d-cysteine during pregnancy can prevent offspring hypertension caused by maternal chronic kidney disease (CKD). The protective effects are related to reducing oxidative stress, modulating the renin-angiotensin system (RAS), and reshaping the gut microbiome.

Perinatal Garlic Oil Supplementation Averts Rat Offspring Hypertension Programmed by Maternal Chronic Kidney Disease

You-Lin Tain et al.

Summary: This study demonstrates that perinatal garlic oil supplementation can protect offspring against hypertension induced by maternal CKD, by modulating H2S signaling, NO, and gut microbiota. The beneficial effects of garlic oil are associated with enhanced H2S signaling, increased NO bioavailability, and shifts in gut microbiota.

Perinatal Propionate Supplementation Protects Adult Male Offspring from Maternal Chronic Kidney Disease-Induced Hypertension

You-Lin Tain et al.

Summary: Emerging evidence suggests that disturbances in early-life gut microbiota can influence adult disease. In this study, the researchers found that maternal chronic kidney disease (CKD) can lead to offspring hypertension, but perinatal supplementation of propionate can prevent this and reverse established hypertension. This protective effect is associated with increased propionate-generating bacteria, elevated plasma propionate levels, higher expression of the SCFA receptor GPR41, improved gut microbiota diversity, and shifts in composition.

Preventing Developmental Origins of Cardiovascular Disease: Hydrogen Sulfide as a Potential Target?

Chien-Ning Hsu et al.

Summary: The cardiovascular system can be programmed by early-life insults, potentially leading to cardiovascular disease in adulthood. Hydrogen sulfide plays a crucial role in the pathogenesis of cardiovascular disease of developmental origins, with early interventions based on H2S showing benefits in preventing adult-onset CVD. Understanding cardiovascular programming and recent advances in H2S-based interventions may help lower the global burden of CVD.

Gut Microbiota Dysbiosis in Human Hypertension: A Systematic Review of Observational Studies

Yang Guo et al.

Summary: This study revealed dysbiosis of gut microbiota in hypertension, characterized by decreased diversity, altered microbial structure, compositional changes in taxa, alterations in microbial function, and stronger microbial interactions. Depletion of short chain fatty acids (SCFAs) producers and over-growth of certain Proteobacteria and Bacteroidetes members were observed in the gut microbiota of hypertensive individuals.
