Tumor microenvironment in glioblastoma: Current and emerging concepts

Pratibha Sharma et al.

Summary: Glioblastoma (GBM) tumor microenvironment (TME) is a highly heterogeneous and complex system, which plays a crucial role in the proliferation, invasion, migration, and survival of GBM cells. Interactions between GBM cells and the TME occur through direct cell-to-cell contact and indirect signaling via cytokines, growth factors, and extracellular vehicles. Recent discoveries of neuron-glioma interfaces and neurotransmitter-based interactions have provided insights into novel mechanisms promoting tumor cell growth and survival. This review discusses the communication between GBM cells and the TME, various models for studying the TME, and current therapeutic strategies targeting TME components.

The Role of Cytokines and Chemokines in Shaping the Immune Microenvironment of Glioblastoma: Implications for Immunotherapy

Erica C. F. Yeo et al.

Summary: Glioblastoma is the most common primary brain tumor in adults, with conventional treatments showing limited improvement in patient survival over the past decade. The immunosuppressive mechanisms employed by tumor cells can hinder treatment efficacy, highlighting the importance of understanding intercellular interactions for potential immunotherapeutic targeting and better patient outcomes.

Tumor Associated Macrophages: Origin, Recruitment, Phenotypic Diversity, and Targeting

Tetiana Hourani et al.

Summary: The tumor microenvironment plays a crucial role in tumorigenesis, with TAMs being closely associated with tumor progression, therapy resistance, and poor survival rates. Targeting TAMs has become an increasingly explored strategy in cancer treatment, with efforts focused on methods like blocking recruitment, stimulating repolarization, and depletion. Additionally, nanomaterials are being investigated for their potential in precise delivery of chemotherapeutic cargo to TAMs.

The Diverse Roles of Heme Oxygenase-1 in Tumor Progression

Kim Ngan Luu Hoang et al.

Summary: HO-1 is an inducible intracellular enzyme expressed in various cancers to promote tumor progression through intracellular and extracellular mechanisms. Its catabolites provide antioxidant, anti-apoptotic, and cytoprotective effects within cells, while also influencing the wider tumor microenvironment to promote processes like angiogenesis, metastasis, anti-inflammation, and immune suppression. Pharmacological inhibition of HO-1 shows promise in inhibiting metastasis and promoting anti-tumor immune responses.

Regulatory effects of IL-1β in the interaction of GBM and tumor-associated monocyte through VCAM-1 and ICAM-1

Ching-Kai Shen et al.

Summary: This study showed that IL-1 beta induces the expression of ICAM-1 and VCAM-1 in GBM cells, leading to increased adhesive interaction between monocytes and GBM cells. Additionally, IL-1 beta also triggers the production of soluble ICAM-1 and VCAM-1 in GBM, which plays a crucial role in regulating tumor-associated monocyte/macrophage polarization.

Macrophage Polarization States in the Tumor Microenvironment

Ava J. Boutilier et al.

Summary: The M1/M2 macrophage paradigm is crucial in tumor progression, where M1 macrophages have anti-tumor effects and M2 macrophages contribute to tumor development through various mechanisms. The tumor microenvironment plays a role in influencing macrophage polarization and activation, which is essential for understanding the pro-tumor effects of tumor-associated macrophages.

Paliperidone Inhibits Glioblastoma Growth in Mouse Brain Tumor Model and Reduces PD-L1 Expression

Yu-Shu Liu et al.

Summary: The study demonstrated that paliperidone inhibits GBM growth and prolongs survival by reducing PD-L1 expression and modulating the interaction between GBM and macrophages through DRD2. The results suggest a potential therapeutic strategy of combining DRD2 antagonists with immunotherapy for the treatment of GBM.
