Identification and validation of oxidative stress and immune-related hub genes in Alzheimer's disease through bioinformatics analysis

Summary: In this study, differentially expressed genes related to oxidative stress in Alzheimer's disease (AD) were identified and analyzed. Immune-related oxidative stress genes and hub genes were further identified and characterized. Functional enrichment analysis revealed the biological functions of these genes. The diagnostic value of 9 key genes was assessed and validated. Additionally, the network relationships among therapeutic agents, miRNAs, and TFs related to the diagnostic genes were established. These findings provide potential therapeutic targets and candidate biomarkers for AD.

Promnesic, Anxiolytic and Antioxidant Effects of Glaucosciadium cordifolium (Boiss.) Burtt & Davis Essential Oil in a Zebrafish Model of Cognitive Impairment

Summary: The aim of this study was to investigate the effects of Glaucosciadium cordifolium essential oil (GCEO, 25 and 150 mu L/L) on anxiety and learning and memory impairment induced by scopolamine (SCOP) in zebrafish. The chemical composition of GCEO was analyzed and the results showed that it mainly consisted of limonene, alpha- and beta-pinene, p-cymene, and alpha-phellandrene. GCEO significantly improved memory impairment and anxiety-like response induced by SCOP through various tests. Furthermore, GCEO reduced oxidative damage and improved cholinergic system activity in the SCOP-induced model.

The Tryptophan-Kynurenine Metabolic System Is Suppressed in Cuprizone-Induced Model of Demyelination Simulating Progressive Multiple Sclerosis

Summary: This study investigated the changes in tryptophan-kynurenine metabolic system in a cuprizone-induced mouse model of demyelination. The results showed a significant reduction in kynurenic acid, 3-hydoxykynurenine, and xanthurenic acid in the plasma, as well as a significant reduction in 3-HK and anthranilic acid in the brain samples during demyelination. These findings suggest that the profile of kynurenine metabolites may serve as a biomarker of progressive multiple sclerosis.

Summary: Anxiety is a serious mental disorder, and around 35.12% of the global population experienced anxiety during the COVID-19 pandemic. Anxiolytic effects can be achieved through the activation of α4β2 nicotinic acetylcholine receptors (nAChRs). Zebrafish, due to its high genetic similarity to humans (70%), has become an ideal animal model for studying anxiety-related behaviors. The novel tank diving test (NTT) is a commonly used paradigm to assess the anxiolytic effects of new drugs. This study found that 3-chloro and 3-bromo derivatives exhibited anxiolytic effects, while chlorine and bromine substitutions at position 5 had an opposite effect.

In silico models also predict that O. onites essential oil may modulate pro-inflammatory enzymes. These findings suggest the therapeutic potential of O. onites essential oil in AD patients.
