Biomineralized Nanoscavenger Abrogates Proinflammatory Macrophage Polarization and Induces Neutrophil Clearance through Reverse Migration during Gouty Arthritis

Summary: A peroxidase-mimicking nanoscavenger was developed to block the secretion of reactive oxygen species (ROS) and proinflammatory cytokines in gouty arthritis induced by monosodium urate (MSU) crystals. The nanoparticles alleviated joint swelling by abrogating ROS and inflammatory cytokine secretions from proinflammatory macrophages, preventing neutrophil infiltration and fluid buildup at the inflammation site. The treatment also reduced macrophage and neutrophil influx in the injured region in zebrafish and mouse models. Overall, the strategy of using biomineralized nanoscavengers shows clinical significance in dual blocking of peroxides and COX2 to prevent inflammation cell influx.

Nanodrug rescues liver fibrosis via synergistic therapy with H2O2 depletion and Saikosaponin b1 sustained release

Summary: Hypoxia and hydrogen peroxide accumulation contribute to liver fibrosis by stimulating hepatic stellate cells (HSCs). Catalase, an antioxidant enzyme, is decreased in liver fibrosis. This study shows that the decrease in catalase is closely related to hypoxia-induced transforming growth factor beta 1 (TGF-beta 1). A nanosystem combining catalase-like MnO2 and anti-fibrosis Saikosaponin b1 (Ssb1) is developed to improve antifibrotic therapy by reducing hypoxic stress and removing excess hydrogen peroxide. This study suggests a promising therapeutic approach for liver fibrosis treatment.

Diagnosis of NAFLD is established through liver biopsy and treatment options include weight loss, dietary modification, and addressing underlying metabolic syndrome.

Role of Oxidative Stress and Lipid Peroxidation in the Pathophysiology of NAFLD

LPO levels above 315.39 μM were identified as an independent risk factor. The model showed good discrimination ability in distinguishing NASH patients. Therefore, understanding the molecular mechanisms of basal inflammation may help identify biomarkers and therapeutic targets for NASH.
