Plasma Levels of sFas-sFasL and FASL Gene Expression Are Associated with Tuberculosis

Iury de Paula Souza et al.

Summary: Apoptosis of macrophages infected by Mycobacterium tuberculosis via Fas-FasL is an important immune mechanism against infection. This study investigated the association between tuberculosis and FAS -670A/G and FASL -124A/G polymorphisms, as well as the levels of sFas and sFasL, and gene expression of FASL and cytokines. The results showed no association between the polymorphisms and tuberculosis. However, the tuberculosis group had higher levels of sFas and lower levels of sFasL. The study also found positive correlations between sFas and sFasL levels, sFasL and FASL expression, and between sFas and FASL expression.

Challenges and the Way forward in Diagnosis and Treatment of Tuberculosis Infection

Kai Ling Chin et al.

Summary: Globally, approximately one-quarter of the world's population is latently infected with Mycobacterium tuberculosis (Mtb), known as latent tuberculosis infection (LTBI). The term tuberculosis infection (TBI) is now used to reflect the dynamic spectrum of the infection, as a small percentage of latent infections progress to active TB. Close contact with index TB patients increases the likelihood of TBI development. The emergence of multidrug-resistant TB (MDR-TB) further complicates the situation. Currently, there is no definitive test for TBI diagnosis, and the available screening tests have limitations in accuracy and predicting disease progression.

Immune checkpoint modulating T cells and NK cells response to Mycobacterium tuberculosis infection

Yongwei Qin et al.

Summary: The down-regulated expression of immune-activating receptors and enhanced expression of immune-inhibitory receptors play a role in chronic infectious diseases and cancer. However, the impact of Mycobacterium tuberculosis infection on the expression of immune checkpoint molecules on natural killer (NK) cells and their functions has been poorly studied. Understanding the function of NK cells during Mtb infection is crucial for a comprehensive understanding of the immune mechanism and evaluation of immunotherapies for treating tuberculosis.

Cytokine storm in tuberculosis and IL-6 involvement

Funmilayo Grace Boni et al.

Summary: Tuberculosis is one of the oldest diseases in humanity, with complications and mortality often associated with cytokine storms. Interleukin-6 (IL-6) plays a crucial role in the immune response to tuberculosis. Researching new therapeutic approaches to block IL-6 signaling is currently a focus in tuberculosis research.

QuantiFERON-TB Gold Plus Assay in Patients With Latent vs. Active Tuberculosis in a Low Incidence Setting: Level of IFN-γ, CD4/CD8 Responses, and Release of IL-2, IP-10, and MIG

Severine Carrere-Kremer et al.

Summary: This study analyzed the relationship between the results of the QuantiFERON Gold Plus assay (QFT) and cytokine patterns associated with active tuberculosis (ATB). The study found that higher levels of IFN-gamma secretion, preferential IFN-gamma response in tube 2, and lower secretion of IL-2, IP-10, and MIG were more likely to be observed in patients with ATB.

Transcriptional Profiling and Deriving a Seven-Gene Signature That Discriminates Active and Latent Tuberculosis: An Integrative Bioinformatics Approach

Sudhakar Natarajan et al.

Summary: This study identified a seven-gene signature with high diagnostic accuracy for discrimination of active and latent tuberculosis. The signature was derived through gene expression analysis of patients from Malawi and South African cohorts, and includes five upregulated genes and two downregulated genes.

Immune evasion and provocation by Mycobacterium tuberculosis

Pallavi Chandra et al.

Summary: In this Review, the authors discuss the strategies used by Mycobacterium tuberculosis to evade the immune response and exploit the host inflammatory response during different phases of its life cycle. They highlight recent studies, identify gaps in knowledge and emphasize the importance of a detailed molecular understanding for the development of novel therapies, vaccines and diagnostics.

Epidemiological and Cytokine Profile of Patients with Pulmonary and Extrapulmonary Tuberculosis in a Population of the Brazilian Amazon

Maria Alice Freitas Queiroz et al.

Summary: Several factors, including epidemiological variables and cytokine levels, are associated with the development of different clinical forms of tuberculosis (TB). This study evaluated these factors in TB patients and controls. It found that older age, marriage, lower education level, and lower family income were associated with TB. Elevated levels of IFN-gamma and IL-4 were more likely in TB patients, and lower IFN-gamma levels were associated with extrapulmonary TB. IFN-gamma levels were found to be useful in differentiating between pulmonary and extrapulmonary TB.

Revisiting tuberculosis screening: An insight to complementary diagnosis and prospective molecular approaches for the recognition of the dormant TB infection in human and cattle hosts

Angel H. Alvarez

Summary: Tuberculosis is a chronic infection in humans and cattle, with many cases remaining subclinical. Diagnostic tests like TST and IGRA are used to detect the infection. While human latent TB infection is recognized, more specific biomarkers are needed to identify dormant infection in cattle.

Oxidative Stress and Inflammatory Mediators in Exhaled Breath Condensate of Patients with Pulmonary Tuberculosis. A Pilot Study with a Biomarker Perspective

Silvia Guzman-Beltran et al.

Summary: Tuberculosis (TB) is a high burden infectious disease worldwide, with inflammation and inadequate antioxidant defense mechanisms believed to contribute to lung damage in TB. This study aimed to evaluate alveolar space-derived metabolites of redox state and inflammation in TB patients and healthy controls using exhaled breath condensate (EBC), finding lower GSH levels and higher levels of inflammatory and oxidant molecules in TB patients' EBC. There was no correlation between EBC and serum, highlighting the importance of measuring local biomarkers for pharmacological and follow-up studies in pulmonary tuberculosis.

Association of IL-4 rs2243250 polymorphism with susceptibility to tuberculosis: A meta-analysis involving 6794 subjects

Summary: The meta-analysis did not find a significant association between IL-4 rs2243250 polymorphism and TB risk overall, but indicated a potential increase in TB risk in the European population with the recessive genetic model. Additional research is needed to clarify the ethnic differences in genetic association studies.
