Possible Side Effects of Polyphenols and Their Interactions with Medicines

Aleksandra Duda-Chodak et al.

Summary: Polyphenols are important components of plant-derived food that have a wide range of beneficial effects on human health. However, they can also have harmful effects, such as blocking iron uptake, inhibiting digestive enzymes, disrupting intestinal microbiota, interacting with drugs, and affecting hormonal balance. Public awareness needs to be raised on the potential side effects of polyphenol supplementation, especially for vulnerable populations.

Gut dysbiosis, defective autophagy and altered immune responses in neurodegenerative diseases: Tales of a vicious cycle

Saravana Babu Chidambaram et al.

Summary: The human microbiota plays a crucial role in regulating various physiological processes and is involved in bidirectional communication with the brain through the Gut-Brain Axis (GBA). Disruptions in the gut microbiota, known as gut dysbiosis, contribute to the pathogenesis of neurodegenerative diseases by triggering chronic neuroinflammation, impaired autophagy, and accumulation of misfolded proteins. Modulating the gut microbiota through interventions such as prebiotics, probiotics, dietary changes, and fecal microbial transplantation (FMT) shows promise as a therapeutic strategy for neurodegenerative diseases.

Multifaceted role of polyphenols in the treatment and management of neurodegenerative diseases

Fahadul Islam et al.

Summary: Neurodegenerative diseases (NDDs) are conditions in which the structure and function of neurons deteriorate over time. While genetic alterations can cause NDDs, there are various physiological systems that can trigger neurodegeneration. Oxidative stress has been linked to several NDDs such as Huntington's, Parkinson's, and Alzheimer's. Synthetic drugs are commonly used to treat NDDs but can have side effects. Studies have discovered natural components in plants that can treat NDDs with lesser side effects, such as polyphenols.

Dietary Polyphenols in Metabolic and Neurodegenerative Diseases: Molecular Targets in Autophagy and Biological Effects

Ana Garcia-Aguilar et al.

Summary: Polyphenols are secondary metabolites of plants that regulate various biological processes, offering protection against inflammation, oxidative stress, and related injuries. They promote autophagy, enhance mitochondrial function and antioxidant defenses, reduce oxidative stress, and decrease cell apoptosis, potentially explaining their health-promoting properties.

Phloretin suppresses neuroinflammation by autophagy-mediated Nrf2 activation in macrophages

Tess Dierckx et al.

Summary: The study shows that phloretin reduces the inflammatory phenotype of macrophages and significantly suppresses neuroinflammation in EAE. Phloretin exerts its effect by activating the Nrf2 signaling pathway, through AMPK-dependent activation of autophagy and subsequent Keap1 degradation. This discovery opens up new perspectives for phloretin as a therapeutic strategy for neuroinflammatory disorders such as MS.

A phenolic-rich extract from Ugni molinae berries reduces abnormal protein aggregation in a cellular model of Huntington's disease

Rodrigo Perez-Arancibia et al.

Summary: The study revealed that extracts from Chilean-native Ugni molinae berries contain anti-inflammatory and antioxidant components, with inhibitory effects on protein aggregation in Alzheimer's and Huntington's disease models. Particularly, extracts from the 19-1 genotype showed significant reduction in polyglutamine peptide aggregation. Additionally, the presence of flavonoids, phenolic acids, and ellagitannins in the extracts was found to be correlated with their protective effects on protein aggregation.

Mitochondrial and Organellar Crosstalk in Parkinson's Disease

Bipul Ray et al.

Summary: Mitochondrial dysfunction is a key factor in Parkinson's disease, affecting protein misfolding and impaired cellular clearance. Dysfunctional communication between mitochondria, endoplasmic reticulum, peroxisomes, and lysosomes may contribute to PD progression. Understanding these interactions could lead to potential therapeutic targets for PD.
