Amyotrophic Lateral Sclerosis Risk Genes and Suppressor

Rupesh Kumar et al.

Summary: Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease characterized by progressive paralysis and respiratory failure. While the disease mechanisms are not fully understood, around 40 genes have been identified as ALS causative genes, with mutations in key genes leading to various cellular dysfunctions. Recent studies have shown that repairing a single cellular pathway can restore other dysfunctions in ALS. This review aims to identify potential therapeutic targets and highlight the importance of a single suppressor in reducing multiple symptoms of ALS.

Infantile SOD1 deficiency syndrome caused by a homozygous SOD1 variant with absence of enzyme activity

Shlomit Ezer et al.

Summary: Pathogenic variants in the SOD1 gene are associated with a severe motor-neurological syndrome in infants, characterized by global developmental delay and movement impairments. This study identified a homozygous loss-of-function variant in the SOD1 gene in an infant with severe neurological symptoms. Further analysis showed that this variant leads to instability and degeneration of the SOD1 protein. The study highlights the importance of specific valine residues in the SOD1 protein and suggests implications for future therapeutic research.

Recent Updates on the Genetics of Amyotrophic Lateral Sclerosis and Frontotemporal Dementia

Laxmi Kirola et al.

Summary: Amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD) are two diseases that primarily affect different areas of the brain, and they share similarities in clinical, genetic, and pathological aspects. Recent research has identified new genes associated with these diseases, and these genes interact with known pathogenic genes, potentially affecting multiple diseases such as Alzheimer's disease, Ataxia, and Parkinsonism. Furthermore, there have been advancements in understanding the genotype-phenotype correlations and in clinical trials for stem cell and gene-based therapies.

Childhood amyotrophic lateral sclerosis caused by excess sphingolipid synthesis

Payam Mohassel et al.

Summary: This study identifies SPTLC1 variants causing excessive sphingolipid biosynthesis as a monogenic form of ALS. Disruption of normal SPT regulation by ORMDL proteins leads to uncontrolled SPT activity and elevated levels of canonical SPT products. Custom-designed small interfering RNAs that selectively target the SPTLC1 ALS allele for degradation offer a potential therapeutic approach to normalize sphingolipid levels.
