Codon pair bias deoptimization of essential genes in infectious laryngotracheitis virus reduces protein expression

Summary: Infectious laryngotracheitis virus (ILTV) is a respiratory pathogen of chickens that causes significant economic losses in the poultry industry and poses health and welfare concerns. This study explored two different codon deoptimization techniques to disrupt and downregulate the expression of essential ILTV genes, ICP8 and UL12. Viruses deoptimized through codon pair bias deoptimization (CPBD) showed decreased protein expression, while viruses deoptimized through codon usage deoptimization (CUD) had inconsistent results. This study demonstrates the potential of CPBD as a tool for studying essential genes in ILTV.

Emergence of a Novel Recombinant Pseudorabies Virus Derived From the Field Virus and Its Attenuated Vaccine in China

Emergence of a novel pathogenic recombinant virus from Bartha vaccine and variant pseudorabies virus in China

Summary: Synonymous recoding of viral genomes by codon-pair deoptimization generates live-attenuated vaccine candidates presumed to be genetically stable, but their actual stability under selective pressure was largely unknown. In this study, a recoded RSV mutant called Min A regained replication fitness and lost its temperature sensitivity through serial passage, acquiring numerous mutations in particular missense mutations in the viral phosphoprotein (P). These P mutations globally increased RSV gene transcription, protein expression, RNA replication, and virus particle production, compensating for the reduced protein expression due to codon-pair deoptimization. Additionally, introduction of the P mutations into Min A generated a genetically stable live-attenuated vaccine candidate with increased attenuation and immunogenicity in hamsters.
